Molecular diagnosis improvement of Roditis leaf discoloration-associated virus causing disease in grapevines

Asma yaprak renk değişikliği ile ilişkili virüs (Grapevine roditis leaf discoloration associated-virus, GRLDaV) Caulimoviridae familyası, Badnavirüs cinsi üyesi olup, endojenik bir virüsdür. Bu çalışmada GRLDaV'nin doğru teşhis için yuvarlanan halka amplifikasyonu (Rolling circle amplification; RCA), virüsün tüm genomunun nükleotid dizisinin elde edilmesi, PCR ve Realtime-PCR temelli teşhis yöntemleri geliştirilmiştir. GRLDaV'un YHA sonucunda amplifikasyonlar edide edilmiş, ancak virüse ait fragmentler bulunamamıştır. Etmen Nicotiana tabacum cv Samsun ve Xanthi ile Chenopodium quinoa test bitkilerine inoküle edilmiş, buradan tekrar tütün bitkisine aktarılmış, PCR analizleri ile teyid edilmiştir. Gen bankasında kayıtlı GRLDaV genomları kullanılarak tasarlanan primerlerle amplifiye edilen 10 genom bölgesinin birleştirilmesi ile tüm GRLDaV B42 izolatının 7,086 nt uzunluğunda tüm genom nükleotid bilgisi elde edilmiştir. Diğer ülkelerde teşhisi yapılan GLRDaV izolatleri ile aynı genom organizasyonuna sahip olup, yüksek oranda nükleotid benzerliği göstermiştir. Sonuç olarak rutin teşhislerde kullanılabilecek primer çiftleri PCR ve real-time PCR analizleri için önerilmiştir.Grapevine leaf discoloration associated virus (GRLDaV) the family of Caulimoviridae is a member of the genus Badnavirus and is an endogenous virus. All genomic nucleotide information of 7,048 nt of all GRLDaV B42 isolates was obtained by combining 10 amplified genome regions with primers designed using GRLDaV genomes registered in the gene bank. It has the same genome organization as the GLRDaV isolates diagnosed in other countries and has shown a high nucleotide similarity.
