Opioids are effective drugs in the treatment of moderate-to-severe pain but development of tolerance, addiction, and severe adverse effects limit their use. The effects of opioids are mediated through opioid receptors that are widely expressed in the central nervous system (CNS) and in the periphery. However, only certain anatomical locations are essential for antinociception and analgesia. Adverse effects may be reduced by targeting opioid exposure to locations important for opioid antinociception and analgesia.
