Antitumor activity of novel plant-derived agents in Medullary Thyroid Carcinoma cells

Das medulläre Schilddrüsenkarzinom (MTC) ist ein neuroendokriner Tumor der parafollikulären C-Zellen der Schilddrüse. Konventionelle Chemotherapeutika zeigen nur geringe Heilungschancen. In der vorliegenden Studie wurden die Wirkungsmechanismen zweier Pflanzen, Cautleya gracilis (Smith) Dandy (CG) und Trailliadoxa gracilis (WW Smith & Forest) (TG), sowie ein Hauptbestandteil von TG, die Ursolsäure (UA), an der humanen MTC-Zelllinie MTC-SK untersucht. Verschiedene Fraktionen von CG, TG und UA wurden in vitro auf morphologische Veränderungen (DAPI und Transmissionsmikroskopie) untersucht, sowie auf Beeinflussung der Proliferation und der Viability (CASY®-1 Cell Counter & Analyzer TTC und WST-1 Cell Proliferation Assay), auf Apoptosevorgänge (Caspasen 3/7, 8 9, 2, 6) sowie Transkriptionsveränderungen an NEMO (real-time PCR). Als normale Kontrolle wurden humane Fibroblasten (HF-SAR) untersucht. Weiters wurden In vivo Versuche mit TG und UA an xenotransplantierten MTC-Mäusen (SCID) gemacht.Fraktionen beider Pflanzen bewirkten in den behandelten Tumorzellen morphologische Veränderungen, Hemmung der Proliferation, der mitochondrialen Aktivität und Vitalität. Gleichzeitig wurde eine gesteigerte Apoptoseaktivität gemessen. Normale Kontrollzellen wurden nicht geschädigt. TG und UA riefen keine Wachstumshemmung der Tumoren in SCID-Mäusen hervor. Auch die NEMO mRNA-Transkripte blieben im Vergleich zu den Kontrollen unverändert. Die vorliegenden Ergebnisse weisen auf eine potentielle Verwendung der untersuchten Wirkstoffe als Chemotherapeutika für MTC hin. Eine frühe Aktivierung der Caspase 8 legte den Verdacht nahe, dass NEMO (NF-?B essential modifier) für die Einleitung der Apoptose verantwortlich sein könnte. Da aber die NEMO mRNA-Transkripte unverändert blieben, muss ein alternativer Weg zum STAT3-Weg zugrunde liegen.Medullary thyroid carcinoma (MTC) is a neuroendocrine tumor arising from the parafollicular C-cells of the thyroid gland which presents poor response to standard chemotherapy. In this study, we analyzed the potential mechanisms of action of two medicinal herbs on a human MTC cell line, MTC-SK. Extracts of Cautleya gracilis (Smith) Dandy (CG) and Trailliaedoxa gracilis (WW Smith & Forrest) (TG) as well as ursolic acid (UA), present in TG, were used to test as potential chemotherapeutic drugs on MTC-SK cells. Normal human fibroblasts (HF-SAR) were used as control. Different fractions of CG, TG and UA were examined in vitro for morphological changes (DAPI staining and transmission electron microscopy), antiproliferative effects (CASY®-1 Cell Counter & Analyzer TTC and WST-1 Cell Proliferation Assay). Apoptosis assays were used to determine the activation of the caspases 3/7, 8, 9, 2 and 6 and real-time PCR (NEMO) was performed to delineate the role of the apoptotic pathway after UA treatment. Additionally, TG and UA were examined in vivo in xenotransplanted MTC-mice (SCID). Morphological changes and antiproliferative effects with inhibition of mitochondrial activity were found in treated tumor cells. Normal control cells were not impaired. Apoptosiswas determined in MTC-SK cells treated with different TG fractions as well as UA but had no change of the NEMO-transcript. No tendency towards tumor inhibition showed the treated MTC-mice.The in vitro-results propose a potential use of the tested agents in antitumor chemotherapy of MTC.
