In the brains of Alzheimer’s disease (AD) patients, two types of lesions coexist: parenchymal deposits formed by amyloid peptide (Aβ) aggregates and intraneuronal aggregation of abnormally hyperphosphorylated Tau proteins. Aβ peptides are derived from the sequential cleavage of the APP protein by two enzymes, β-secretase and γ-secretase. The vast majority of molecules in clinical trials currently targets one or the other of the two pathologies, and AD is defined by the co-existence of these two lesions. Thus, several studies suggest a synergistic link between the two lesional processes, and a treatment acting on both amyloid pathology and Tau pathology would then appear as a highly interesting therapeutic strategy.
