Reverse transcription of human immunodeficiency virus typa-1 (HIV-1),study of various initiation complexe and mechanism of resistance toward nucleoside reverse transcriptase inhibitors (NRTIs)

La rétrotranscription de HIV-1 permet la synthèse de l'ADN proviral bicaténaire à partir de l'ARN génomique. Elle requiert la présence de l'ARN viral (ARNv) qui sert de matrice, l'ARNt3Lys cellulaire qui sert d'amorce, et la rétrotranscriptase. L'extrémité 3 de l'ARNt3Lys est strictement complémentaire d'une séquence appelée Primer Binding Site où débute la rétrotranscription. Outre cette interaction, des remaniements conformationnels intra- et intermoléculaires ont lieu lors de la formation du complexe ARNt3Lys/ARNv de l'isolat MAL'de HIV-1. Nous avons identifié les éléments structuraux du complexe d'initiation de l'isolat MAL'de HIV-1 cruciaux pour l'initiation de la rétrotranscription. Nos résultats montrent le rôle essentiel de la structure tridimensionnelle globale dans le processus d'initiation. Puis, nous avons entrepris une étude structurale (in vitro et in situ) et fonctionnelle comparative de deux isolats, MAL'et Hxb2, dont les domaines PBS appartiennent respectivement aux sous-types A et B. Cette étude a montré la versatilité du système HIV-1. Contrairement à la situation observée avec l'isolat MAL, l'interaction entre le PBS de l'ARNv de l'isolat Hxb2 et l'extrémité 3 de l'ARNt3Lys est suffisante pour initier la rétrotranscription, en absence de tout remaniement conformationnel.Lors du traitement par l'AZT, de nombreuses mutations de résistance apparaissent. Bien que les mutations de résistance permettent une réparation par ATP-lyse, la RT sauvage présente une résistance innée via le mécanisme de pyrophosphorolyse, dont l'efficacité d'enlèvement est supérieure à celle obtenue par ATP-lyse en présence de l'enzyme résistante.La concentration en Mg2+ libre influence les activités RNase H et polymérase de la RT de HIV-1, en agissant sur l'incorporation de nt et la processivité de l'enzyme, ainsi que la stabilité de la matrice ARN. De plus, l'inhibition de la synthèse d'ADN par un terminateur de chaîne est dépendante de la concentration en Mg2+.[...]Reverse transcription of HIV-1 genomic RNA is primed by tRNA3Lys, whose 3' end 18 nucleotides are complementary to the viral primer binding site (PBS). However, in the HIV-1 MAL isolate, additional interactions between tRNA3Lys and the genomic RNA occur during formation of the initiation complex.We studied the role of the different structural elements of the initiation complex of the HIV-1 MAL isolate and showed that the overall 3D structure of the complex is required for efficient initiation of reverse transcription. We also compared the initiation complexes formed by two highly divergent isolates (MAL and HXB2) by performing in situ structural probing as well as in vitro structural and functional studies. Surprisingly, our results showed that the structure of the initiation complex is not conserved and therefore highlighted the versatility of the HIV-1 system. Indeed, in the MAL isolate and, according to sequence analysis, in 14% of all HIV-1 isolates, formation of the initiation complex requires complex rearrangements of the viral RNA and extensive interactions with tRNA3Lys. In contrast, in the HXB2 isolate, as well as in most isolates, tRNA3Lys annealing only minimally affects the viral RNA structure and no interaction outside the PBS is required. AZT is a widely used inhibitor of HIV-1 reverse transcriptase (RT) that acts as a chain terminator. However rapid emergence of multiresistant HIV-1 strains appears. It was believed that resistance toward AZTTP is due to the increased unblocking, by the AZT-resistant RT, of the AZT-terminated primer via ATP-lysis. However, detailed comparisons performed with the wild type and resistant enzymes indicated that pyrophosphorolysis is more efficient than the ATP-mediated unblocking.Both the polymerase and RNase H activities of HIV-1 RT are strongly affected by the Mg2+ concentration.
