UJI AKTIVITAS ANTIBAKTERI FRAKSI N-HEKSAN DAUN SIRIH HIJAU (Piper betle L.) TERHADAP PERTUMBUHAN BAKTERI Propionibacterium acnes SECARA IN VITRO

Background: Green betel leaf (Piper betle L.) is one of the medicinal plants that has been widely studied for its properties. The potential of this plant, among others, is as an antiseptic, antibacterial, antioxidant, and fungicide. The betel plant is used as a deodorant, mouthwash, eliminating vaginal discharge, digestive disorders, and various infections (Hoque et al., 2011). The active compounds that have antibacterial properties contained in Piper betle L. are alkaloids, flavonoids, polyphenols, and essential oils. Objectives: Determine the antibacterial activity and profile of secondary metabolites contained in the n-hexane fraction of green betel leaf (Piper betle L.) against Propionibacterium acnes bacteria. Method: Identification of compound groups contained using the thin layer chromatography (TLC) method. Kirby Bauer method (disk diffusion) is the method applied to the antibacterial activity test. Result and Conclusion: The results of the antibacterial activity test showed that the diameter of the inhibition zone was as follows: a concentration of 500 mg/ml of 10.23 mm; a concentration of 250 mg/ml of 10.03 mm; and at a concentration of 125 mg/ml of 9.6 mm. Identification of the n-hexane fraction of green betel leaf detected terpenoids, alkaloids, polyphenols, anthraquinones, and flavonoids. So this study concludes that the n-hexane fraction of green betel leaf (Piper betle L.) is proven to be able to inhibit the growth of Propionibacterium acnes bacteria, and this fraction contains a class of compounds that have antibacterial activity, including terpenoids, alkaloids, polyphenols, anthraquinones, and flavonoids
