New analytical methodologies for the detection of residues and metabolites of pharmaceuticals in the marine environment of fish farms

The rapid urbanization of free land, industrial progress and the improvement of the quality of human life have led to the development and increased consumption of various chemicals that are recognized as environmental pollutants. So-called "emerging contaminants" are chemicals that pose a potential risk to both the environment and human health, according to the US EPA and their concentrations range from ng L-1 to μg L-1. This category includes pharmaceutical substances, due to their widespread use, their physicochemical properties and their incomplete removal from wastewater treatment plants (WWTPs).A group of such substances widely used are pharmaceutical substances and personal care products (Pharmaceuticals and Personal Care Products, PPCPs), intended for human and veterinary use. In addition to prescription drugs, these products also include diagnostic and nutritional substances, perfumes, sunscreens, a number of cosmetics and others. Medicinal substances and their active metabolites constantly enter the aquatic environment, as complex mixtures, through various routes. Their residues have been detected both in groundwater and surface water, as well as in municipal and hospital wastewater.The main sources of these pharmaceutical substances in the environment are urban wastewater treatment plants, agricultural facilities, aquaculture, hospitals and drug production facilities. Therefore, in the last 15 years substantial research has been carried out to determine the presence, fate, effects and risks of these pharmaceutical substances in the environment.At national level, research on the effects on coastal areas is limited, while even fewer are the surveys concerning areas with a high activity of aquaculture, which are a significant potential source of marine pollution. In order to reach a safe conclusion about the pollution of an ecosystem, it is imperative need the analysis of other substrates besides aqueous samples and other substrates such as sediments and plant-animal organisms.Although Greece is considered a small European country, it is ranked among the countries with the highest consumption of pharmaceutical substances per person, after the United States and Canada. Nevertheless, the actual percentage of drugs used is inaccurate and significantly higher for some substances, taking into account the fact that the annual consumption of a drug is not easy to determine and is often based on estimates.Based on the above, the purpose of the thesis is initially: (a) the development, optimization and validation of an analytical method for the determination of residues of pharmaceutical compounds in water, sediment and fish of fish farms, as well as the systematic control for the determination of the concentration levels of the residues of the selected pharmaceutical compounds and their metabolites in the above substrates over an annual monitoring period. Followed by the selection of two characteristic representatives of pharmaceutical substances (antibiotics sulfathiazole and sulfamethoxypyridazine) for further research of their behavior, where it was carried out: (b) a study of their photolytic degradation in different types of surface water (river, lake, seawater, distilled) both in environmental and laboratory conditions, where the following were studied: kinetics, determination of transformation products, toxicity study, determination of inorganic ions and total organic carbon.A simple analytical method for the determination of pharmaceuticals in aqueous samples of aquaculture was developed, optimized and validated. The method was based on the application of solid phase extraction (SPE). Regarding the other two substrates studied (sediment, fish), a simple, fast, economic and environmentally friendly method, was developed, optimized and validated, the multi-residue method QuEChERS (quick, easy, cheap, effective, rugged, safe). The qualitative and quantitative determination of the selected pharmaceutical compounds was based on the use of ultra-high performance liquid chromatography (UHPLC) coupled with mass spectrometry of high resolution and accuracy (high-resolution-accurate-mass spectrometry, HRMS) and more specifically with the hybrid analyzer LTQ/ Orbitrap MS.A total of 21 active pharmaceutical substances were selected (citalopram, venlafaxine, fluoxetine hydrochloride, erythromicin, carbamazepine, sulfadiazine, sulfamethazine, sulfamethizole, sulfamethoxazole, sulfamethoxypyridazine, sertraline hydrochloride, sulfapyridine, sulfaquionoxaline, sulfathiazole, caffeine, paracetamol, trimethoprim, carbamazepine, phenazone, oxytetracycline, oxolinic acid) and 5 metabolites (O-desmethyl venlafaxine, norfluoxetine hydrochloride, N-acetylsulfamethoxazole, carbamazepine-10,11-epoxide, N-desmethyl sertraline hydrochloride). The selection of these pharmaceutical compounds was based on their high annual consumption and detection in surface waters, as well as the relative concern about their potential effects on both human and aquatic organisms. The samples used for the development of analytical methods for the substrates studied (seawater, fish, sediment) as well as the real samples used by the optimized and validated methods were collected from a fish farm located in the region of Epirus (Ionian Sea). Annual systematic testing was carried out for the samples of seawater and fish (sea bream, S.aurata), from July 2020 to June 2021, while the real sediment samples collected from the fish farm and seasonal sampling was carried out, covering the four seasons: autumn 2020 (September), winter 2021 (February), spring 2021 (May) and summer 2021 (June).The suitability of the SPE method for the determination of the selected pharmaceuticals in seawater was confirmed by the analytical characteristics of the method. Recoveries for the midfield ranged from 64.8% to 115.4%, for the high level they were found between 62.4% and 118.8% and for the low level from 61.6% to 98.2%. The repeatability of the method (RSDr) for the low level ranged from 0.6% to 1.8%, for the middle level from 0.5% to 3.9% and for the high level the values ranged from 0.5% to 2.2%. The reproducibility (RSDR) for the small level ranged from 1.1% to 3.2%, for the median level from 1.1% to 3.1% and for the high level from 0.9% to 4.8%. . Detection limits ranged from 0.3 ng L-1 to 9.8 ng L-1, while limits of quantification ranged from 1.2 ng L-1 to 26.4 ng L-1.The optimized analytical method was applied to real samples with a twelve-month monitoring study (July 2020 - June 2021). Positive detections were found for the substances oxytetracycline with a maximum concentration of 43817 ng/L (February '21), sulfadiazine 1092 ng/L (February '21), trimethoprim with a concentration of 34.2 ng/L (May '21), paracetamol 343.5 ng/L (December '20) and caffeine with a maximum concentration of 59.7 ng/L (December '20).The suitability of the multi-residue QuEChERS method for the determination of the selected compounds in fish samples (sea bream) was confirmed by the analytical characteristics of the method. Recoveries for the low level ranged from 61.8% to 99.2%, recoveries for the midfield were found between 64.0% to 107.0%, while for the high level from 64.4% to 103.2%. The repeatability of the method (RSDr) for the small level ranged from 0.6% to 2.9%, for the middle level from 0.5% to 3.9% and for the high level the values ranged from 0.5% and 3.7%. The within-laboratory reproducibility (RSDR) for the small level ranged from 1.2% to 5.5%, for the intermediate level from 2.1% to 6.2% and for the high level from 0.9% to 6.9%. Limits of detection ranged from 0.3 to 7.0 ng g-1, while limits of quantification ranged from 0.5 to 19.0 ng g-1.The optimal analytical methodology developed was applied to real samples with a monitoring study lasting twelve months (July 2020-June 2021). Positive detection was found for the antibiotic trimethoprim 26.4 ng/g (November '20).The suitability of the QuEChERS method for the determination of the selected compounds in the sediment samples was confirmed by the analytical characteristics of the method. Recoveries for the low level ranged from 60.4% to 95.4%, recoveries for the midfield were found between 63.0% to 96.2%, while for the high level from 64.0% to 99.2%. The repeatability of the method (RSDr) for the small level ranged from 0.8% to 2.5%, for the middle level from 0.6% to 2.8% and for the high level the values ranged from 0.7% to 2.2%. The within laboratory reproducibility (RSDR) for the small level ranged from 1.1% to 4.2%, for the intermediate level from 1.6% to 4.2% and for the high level from 1.0% to 5.8%. Detection limits ranged from 0.4 ng g-1 to 12.3 ng g-1, while quantification limits were found from 1.5 ng g-1 to 35.5 ng g-1.The analytical methodology developed was applied to real sediment samples from fish farming in seasonal sampling (one sample was analyzed for each season). None of the selected pharmaceuticals were detected besides caffeine, which was also found at a concentration below the quantification limit.The risk assessment (acute and chronic) was carried out using the risk quotient method and the sum of toxic units for the three trophic levels (fish, invertebrates, algae). From the calculations of the risk quotient of acute toxicity, more serious pressures were found for the taxonomic groups of invertebrates and algea. In particular, high toxicity exhibited paracetamol and sulfadiazine compounds in invertebrates and algae, respectively (RQ> 1), as well as the antibiotic oxytetracycline, which exhibited high acute toxicity in all three taxonomic groups. The calculation of risk quotient for chronic toxicity gave a high risk for oxytetracycline in invertebrates and algea.Moreover, a study of the photolytical degradation of the two selected organic compounds (sulfathiazole and sulfamethoxypyridazine) was carried out. The selection of two antibiotics was mainly based on their wide and extensive use worldwide in recent years and their high rates of detection in surface waters.The study of the photolytical degradation of the two pharmaceuticals was carried out in distilled water and three different types of surface waters (river, lake, seawater), under the influence of both an artificial (suntest solar radiation), and natural solar radiation. In both cases, the photolysis of the surface waters was faster when it was carried out under the artificial solar radiation than under natural solar radiation. However, the two antibiotics presented in both cases a lower pace of photolytic degradation in distilled water rather than in surface waters, indicating that the presence of dissolved organic matter in surface waters lead to an increase in the photolytic degradation of the selected antibiotics. Moreover, the use of humic acids and nitrate ions as photosynthetizers inhibit the indirect photodegradation of sulfathiazole and sulfamethoxypyridazine. The degradation of the two substances presented a pseudo-first-order kinetics. Identification of the intermediate products was performed using UHPLC/LTQ-Orbitrap, where 4 transformation products were determined for sulfathiazole and 7 transformation products for sulfamethoxypyridazine. At the same time, photolytic experiments were performed using photosensitizers in distilled water and in the three different surface waters (river, lake, seawater), under the effect of artificial solar radiation. Humic acids and nitrates were used as photosensitizers at concentrations of 2.5, 5, 10, 20 mg/l, respectively. After identifying and monitoring the kinetics and degradation of the transformation products of the two pharmaceuticals, their toxicity was studied using the Microtox Test system (Microtox 500, AZUR), using Vibrio Fischeri bacteria.For the determination of anions (fluoride, chloride, bromide, nitrate, nitrite and sulfate), LC-10AD VP liquid chromatography coupled with CDD-6A conductivity detector and anionic column IC-A3 of Shimadzu was used. In order to estimate the percentage of mineralization and oxidation of the two selected pharmaceuticals, the calculation of Total Organic Carbon, (TOC), was performed at the Shimadzu TOC-L-Series analyzer.Η ταχεία αστικοποίηση της ελεύθερης γης, η βιομηχανική πρόοδος και η βελτίωση της ποιότητας της ανθρώπινης ζωής οδήγησε στην ανάπτυξη και την αυξημένη κατανάλωση διαφόρων χημικών ουσιών που αναγνωρίζονται ως περιβαλλοντικοί ρυπαντές. Οι αποκαλούμενοι ως «αναδυόμενοι ρύποι» (emerging contaminants) είναι χημικές ουσίες, οι οποίες αποτελούν πιθανό κίνδυνο τόσο για το περιβάλλον όσο και για την ανθρώπινη υγεία, σύμφωνα με την υπηρεσία περιβάλλοντος των ΗΠΑ (US EPA – Environmental Protection Agency) και βρίσκονται σε εύρος συγκεντρώσεων από ng L-1 έως μg L-1. Στην κατηγορία αυτή, ανήκουν οι φαρμακευτικές ουσίες, εξαιτίας της ευρείας χρήσης τους, των φυσικοχημικών ιδιοτήτων τους και της ατελούς απομάκρυνσής τους από τις μονάδες επεξεργασίας λυμάτων (wastewater treatment plants, WWTPs). Μία ομάδα τέτοιων ουσιών ευρέως χρησιμοποιούμενων αποτελούν οι φαρμακευτικές ουσίες και τα προϊόντα προσωπικής φροντίδας (Pharmaceuticals and Personal Care Products, PPCPs), που προορίζονται για ανθρώπινη και κτηνιατρική χρήση. Εκτός από τα συνταγογραφούμενα φάρμακα, στα προϊόντα αυτά ανήκουν επίσης οι διαγνωστικές και θρεπτικές ουσίες, τα αρώματα, τα αντηλιακά, πλήθος καλλυντικών και άλλα. Οι φαρμακευτικές ουσίες και οι ενεργοί μεταβολίτες τους εισέρχονται συνεχώς στο υδάτινο περιβάλλον, ως πολύπλοκα μίγματα, μέσω διαφόρων οδών. Έχουν ανιχνευτεί υπολείμματά τους τόσο στα υπόγεια και επιφανειακά ύδατα, όσο και στα αστικά και νοσοκομειακά υγρά απόβλητα.Οι κυριότερες πηγές των φαρμακευτικών αυτών ουσιών στο περιβάλλον είναι οι αστικές μονάδες επεξεργασίας λυμάτων, γεωργικές εγκαταστάσεις, υδατοκαλλιέργειες, νοσοκομεία και εγκαταστάσεις παραγωγής φαρμάκων. Ως εκ τούτου, τα τελευταία 15 χρόνια έχει πραγματοποιηθεί ουσιαστική έρευνα για να προσδιοριστεί η παρουσία, η τύχη, τα αποτελέσματα και οι κίνδυνοι των φαρμακευτικών αυτών ουσιών στο περιβάλλον. Σε εθνικό επίπεδο, οι έρευνες που αφορούν τις επιπτώσεις σε παράκτιες περιοχές είναι περιορισμένες, ενώ ακόμα λιγότερες είναι οι έρευνες που αφορούν περιοχές με έντονη δραστηριότητα υδατοκαλλιεργειών, που αποτελούν σημαντική εν δυνάμει πηγή θαλάσσιας ρύπανσης. Προκειμένου να εξαχθεί ένα ασφαλές συμπέρασμα για τη ρύπανση ενός οικοσυστήματος είναι επιτακτική ανάγκη η ανάλυση εκτός των υδατικών δειγμάτων και άλλων υποστρωμάτων όπως τα ιζήματα και οι φυτικοί-ζωικοί οργανισμοί.Παρότι η Ελλάδα θεωρείται μια μικρή Ευρωπαϊκή χώρα, κατατάσσεται στις χώρες με την υψηλότερη κατανάλωση φαρμακευτικών ουσιών κατ’ άτομο, μετά τις Ηνωμένες Πολιτείες και τον Καναδά. Παρόλα αυτά βέβαια, το πραγματικό ποσοστό των χρησιμοποιούμενων φαρμάκων είναι ανακριβές και σημαντικά υψηλότερο για κάποιες ουσίες, λαμβάνοντας υπόψη το γεγονός ότι η ετήσια κατανάλωση ενός φαρμάκου δεν είναι εύκολο να προσδιοριστεί και συχνά βασίζεται σε εκτιμήσεις.Με βάση τα παραπάνω, σκοπός της διατριβής είναι αρχικά: (α) η ανάπτυξη, η βελτιστοποίηση και η επικύρωση αναλυτικής μεθόδου για τον προσδιορισμό υπολειμμάτων φαρμακευτικών ενώσεων σε νερό, ίζημα και ψάρια ιχθυοκαλλιεργειών, καθώς και ο συστηματικός έλεγχος για τον προσδιορισμό των επιπέδων συγκέντρωσης των υπολειμμάτων των επιλεγμένων φαρμακευτικών ενώσεων και των μεταβολιτών τους στα ανωτέρω υποστρώματα σε διάστημα ετήσιας παρακολούθησης (monitoring). Ακολούθησε η επιλογή δύο χαρακτηριστικών εκπροσώπων των φαρμακευτικών ουσιών (των αντιβιοτικών sulfathiazole και sulfamethoxypyridazine) για περεταίρω έρευνα της συμπεριφοράς τους, όπου πραγματοποιήθηκε: (β) μελέτη της φωτολυτικής τους αποικοδόμησης σε διαφορετικούς τύπους επιφανειακών υδάτων (ποτάμιο, λιμναίο, θαλάσσιο, απεσταγμένο) τόσο σε συνθήκες περιβάλλοντος όσο και σε συνθήκες εργαστηρίου, όπου μελετήθηκαν τα εξής: κινητική, προσδιορισμός προϊόντων μετασχηματισμού, μελέτη τοξικότητας, προσδιορισμός ανόργανων ιόντων και ολικού οργανικού άνθρακα.Αναπτύχθηκε, βελτιστοποιήθηκε και επικυρώθηκε μια απλή αναλυτική μέθοδος προσδιορισμού των φαρμακευτικών ουσιών στα υδατικά δείγματα ιχθυοκαλλιέργειας. Η μέθοδος βασίστηκε στην εφαρμογή της εκχύλισης δια της στερεάς φάσης (Solid Phase Extraction, SPE). Όσον αφορά τα άλλα δύο υποστρώματα που μελετήθηκαν (ίζημα, ψάρια) αναπτύχθηκε, βελτιστοποιήθηκε και επικυρώθηκε μία απλή, γρήγορη, οικονομική και φιλική προς το περιβάλλον μέθοδος, η πολυ-υπολειμματική μέθοδος QuEChERS (quick, easy, cheap, effective, rugged, safe). Ο ποιοτικός και ποσοτικός προσδιορισμός των επιλεγμένων φαρμακευτικών ενώσεων βασίστηκε στη χρήση υγρής χρωματογραφίας υπερ-υψηλής απόδοσης (ultra-high performance liquid chromatography, UHPLC) συζευγμένο με φασματομετρία μάζας υψηλής διακριτικής ικάνοτητας και ακρίβειας (high-resolution-accurate-mass spectrometry, HRMS) και πιο συγκεκριμένα με τον υβριδικό αναλυτή LTQ/Orbitrap MS.Επιλέχθηκαν συνολικά 21 δραστικές φαρμακευτικές ουσίες (citalopram, venlafaxine, fluoxetine hydrochloride, erythromicin, carbamazepine, sulfadiazine, sulfamethazine, sulfamethizole, sulfamethoxazole, sulfamethoxypyridazine, sertraline hydrochloride, sulfapyridine, sulfaquionoxaline, sulfathiazole, caffeine, paracetamol, trimethoprim, carbamazepine, phenazone, oxytetracycline, oxolinic acid) και 5 προϊόντα μετασχηματισμού (O-desmethyl venlafaxine, norfluoxetine hydrochloride, N-acetylsulfamethoxazole, carbamazepine-10,11-epoxide, N-desmethyl sertraline hydrochloride). Η επιλογή των φαρμακευτικών αυτών ενώσεων βασίστηκε στην υψηλή ετήσια κατανάλωσή τους και στην ανίχνευσή τους στα επιφανειακά ύδατα, καθώς επίσης και τη σχετική ανησυχία που υπάρχει για τις πιθανές επιπτώσεις τους τόσο στους ανθρώπινους όσο και στους υδάτινους οργανισμούς. Τα δείγματα που χρησιμοποιήθηκαν για την ανάπτυξη των αναλυτικών μεθόδων για τα υποστρώματα που μελετήθηκαν (θαλασσινό νερό, ψάρι, ίζημα) καθώς και τα πραγματικά δείγματα που εφαρμόστηκαν οι βελτιστοποιημένες και επικυρωμένες μέθοδοι συλλέχθηκαν από ιχθυοκαλλιέργεια που βρίσκεται στην περιοχή της Ηπείρου (Ιόνιο Πέλαγος). Πραγματοποιήθηκε ετήσιος συστηματικός έλεγχος για τα δείγματα του θαλασσινού νερού και των ψαριών (τσιπούρας, S.aurata), από τον Ιούλιο 2020 μέχρι τον Ιούνιο 2021, ενώ για τα πραγματικά δείγματα ιζήματος από ιχθυοκαλλιέργεια πραγματοποιήθηκε εποχιακή δειγματοληψία, καλύπτοντας τις τέσσερις εποχές: φθινόπωρο 2020 (Σεπτέμβριος), χειμώνας 2021 (Φεβρουάριος), άνοιξη 2021 (Μάιος) και καλοκαίρι 2021 (Ιούνιος).Η καταλληλότητα της SPE για τον προσδιορισμό των επιλεγμένων ενώσεων στο θαλασσινό νερό επιβεβαιώθηκε από τα αναλυτικά χαρακτηριστικά της μεθόδου. Οι ανακτήσεις για το μεσαίο επίπεδο κυμάνθηκαν από 64.8% έως 115.4%, για το υψηλό επίπεδο βρέθηκαν μεταξύ 62.4% και 118.8% και για το χαμηλό επίπεδο από 61.6% έως 98.2%. Η επαναληψιμότητα της μεθόδου (RSDr) για το μικρό επίπεδο κυμάνθηκε από 0.6% έως 1.8%, για το μεσαίο επίπεδο από 0.5% έως 3.9% και για το υψηλό επίπεδο οι τιμές κυμάνθηκαν από 0.5% έως 2.2%. Η ενδοεργαστηριακή αναπαραγωγιμότητα (RSDR) για το μικρό επίπεδο κυμάνθηκε από 1.1% έως 3.2%, για το μεσαίο επίπεδο από 1.1% έως 3.1% και για το υψηλό επίπεδο από 0.9% έως 4.8%. . Τα όρια ανίχνευσης κυμάνθηκαν από 0.3 ng L-1 έως 9.8 ng L-1, ενώ τα όρια ποσοτικοποίησης βρέθηκαν από 1.2 ng L-1 έως 26.4 ng L-1.Η βελτιστοποιημένη αναλυτική μέθοδος που αναπτύχθηκε εφαρμόστηκε σε πραγματικά δείγματα με μελέτη παρακολούθησης (monitoring study) διάρκειας δώδεκα μηνών (Ιούλιος 2020- Ιούνιος 2021). Θετικές ανιχνεύσεις βρέθηκαν για τις ουσίες oxytetracycline με μέγιστη συγκέντρωση 43817 ng/L (Φεβρουάριος ’21), sulfadiazine 1092 ng/L (Φεβρουάριος ’21), trimethoprim με συγκέντρωση 34.2 ng/L (Μάϊος ’21), paracetamol 343.5 ng/L (Δεκέμβριος ’20) και caffeine με μέγιστη συγκέντρωση 59.7 ng/L (Δεκέμβριος ’20). Η καταλληλότητα της πολυ-υπολειμματικής μεθόδου QuEChERS για τον προσδιορισμό των επιλεγμένων ενώσεων στα δείγματα ψαριού (τσιπούρα) επιβεβαιώθηκε από τα αναλυτικά χαρακτηριστικά της μεθόδου. Οι ανακτήσεις για το χαμηλό επίπεδο κυμάνθηκαν από 61.8% έως 99.2%, οι ανακτήσεις για το μεσαίο επίπεδο βρέθηκαν μεταξύ 64.0% έως 107.0%, ενώ για το υψηλό επίπεδο από 64.4% έως 103.2%. Η επαναληψιμότητα της μεθόδου (RSDr) για το μικρό επίπεδο κυμάνθηκε από 0.6% έως 2.9%, για το μεσαίο επίπεδο από 0.5% έως 3.9% και για το υψηλό επίπεδο οι τιμές κυμάνθηκαν από 0.5% και 3.7%. Θετική ανίχνευση βρέθηκε για το αντιβιοτικό trimethoprim 26.4 ng/g (Νοέμβριος ’20). Η καταλληλότητα της μεθόδου QuEChERS για τον προσδιορισμό των επιλεγμένων ενώσεων στα δείγματα ιζήματος επιβεβαιώθηκε από τα αναλυτικά χαρακτηριστικά της μεθόδου. Οι ανακτήσεις για το χαμηλό επίπεδο κυμάνθηκαν από 60.4% έως 95.4%, οι ανακτήσεις για το μεσαίο επίπεδο βρέθηκαν μεταξύ 63.0% έως 96.2%, ενώ για το υψηλό επίπεδο από 64.0% έως 99.2%.
