Chemical study, toxicological and neuroprotective effect of garlic passion fruit (Passiflora tenuifila Killip)

Several species of the genus Passiflora are its sedative and soothing properties. Passiflora tenuifila is a type of non-commercial passion fruit known as garlic passion fruit and is among the least known, with few scientific studies in the literature with their different properties. The aim was to characterize the ingredients present in passion fruit and to evaluate the toxicity and effects on the Central Nervous System (CNS) in the animal models. The fruits were ground whole and lyophilized. After, were characterized in the physical-chemical and phytochemical analytics. Toxicity was evaluated in in vitro and in vivo models. Anti-inflammatory activity was evaluated in vitro with human neutrophils. Anxiolytic, antidepressive, sedative and anticonvulsant effects were evaluated in a behavioral model using Swiss mice and the evaluation of the antiparkinsonian effect was performed with Wistar rats using rotenone as an inducing agent for Parkinson's symptoms. Garlic passion fruit presented a moisture content of 4.99%, 6.29% of ash, 8.77% of proteins, 10.93% of lipids and 69.19% of carbohydrates, of these 48.40% correspond to the content of total dietary fiber. Regarding the total polyphenol content and total antioxidant activity, 1151.34 mg EAG.100 g-1 and 109.71 μM Trolox.g-1, respectively. In the chemical composition by Nuclear Magnetic Resonance (NMR) were identified sugars, organic acids and amino acids and by Ultra-High Performance Liquid Chromatography (UPLC), phenolic compounds, mainly phenolic acids and flavonoids. There was no in vitro toxicity and in vivo tests showed that the administrations of garlic passion fruit used did not cause any signs of toxicity in relation to the evaluated parameters. Anti-inflammatory activity was no observed in human neutrophils at the doses used. In the functional effect, the animals treated with garlic passion fruit (200 and 400 mg.kg-1) showed a decrease in the locomotor activity in the open field test and without alterations in the motor coordination in the test of the routed rod, indicating a sedative activity without muscle relaxation. In addition, they also showed an increase in the behavior of head dips in the hole board test, indicating an anxiolytic activity. The forced swimming test was performed to assess antidepressant activity, but no significant effect was observed at the doses used. Garlic passion fruit (200 and 400 mg.kg-1) potentiated sleep induced by ethyl ether, showing a hypnotic-sedative activity. In the seizure test induced by pentylenetetrazol, it was observed that the garlic passion fruit (400 mg.kg-1) exerted a protective effect, reducing the severity of the seizures and not causing the death of the animals. In the anti-parkinsonian effect, behavioral tests indicated that consumption of garlic passion fruit caused an improvement and possible recovery effect of rotenone-induced damage in the CNS, and in addition, caused an increase in the level of dopamine in the brains of animals in the dose of 400 mg.kg-1. It can be concluded that garlic passion fruit presented no toxicity, being safe for human consumption, and presented promising functional effect as anxiolytic agent, hypnotic-sedative, anticonvulsive and antiparkinsonian, possibly related to the phenolic compounds present.Diversas espécies do gênero Passiflora são conhecidas por suas propriedades sedativas e calmantes. Passiflora tenuifila Killip é uma espécie de maracujá não comercial, conhecida por maracujá alho e está entre as menos conhecidas, existindo poucos estudos científicos na literatura relatando suas propriedades funcionais. Este trabalho teve por objetivo caracterizar os componentes químicos presentes nos frutos de maracujá alho e avaliar a toxicidade e os efeitos funcionais no Sistema Nervoso Central (SNC) em modelos animais. Os frutos foram triturados inteiros e liofilizados. Para caracterização foram realizadas análises físicos-químicas e fitoquímicas. A toxicidade foi avaliada em modelos in vitro e in vivo. A atividade anti-inflamatória foi avaliada in vitro com neutrófilos humanos. A avaliação do efeito ansiolítico, antidepressivo, sedativo e anticonvulsivante foi realizada em modelo comportamental utilizando camundongos Swiss e a avaliação do efeito antiparkinsoniano foi realizada com ratos Wistar, utilizando rotenona como agente indutor dos sintomas de Parkinson. O maracujá alho apresentou teor umidade de 4,99%, 6,29% de cinzas, 8,77% de proteínas, 10,93% de lipídios e de 69,19% de carboidratos, destes 48,40% correspondem ao conteúdo de fibra alimentar total. Em relação ao conteúdo de polifenóis totais e atividade antioxidante total apresentou 1151,34 mg EAG/100 g e 109,71 µM Trolox/g, respectivamente. Na avaliação da composição química por Ressonância Magnética Nuclear (RMN) foram identificados açúcares, ácidos orgânicos e aminoácidos e por Cromatrografia Líquida de Ultra Eficiência (UPLC) compostos fenólicos, principalmente ácidos fenólicos e flavonoides. Não apresentou toxicidade in vitro e os testes in vivo evidenciaram que as concentrações de maracujá alho testadas não causaram nenhum sinal de toxicidade. Não foi observada atividade anti-inflamatória em neutrófilos humanos nas doses utilizadas. Na avaliação de efeito funcional, os animais tratados com maracujá alho (200 e 400 mg/kg) apresentaram uma diminuição na atividade locomotora no teste do campo aberto e sem alterações na coordenação motora no teste do rota rod, indicando uma ação sedativa sem relaxamento muscular. Além disso, houve aumento no comportamento de head dips no teste da placa perfurada, indicando uma atividade ansiolítica. O teste do nado forçado foi realizado para avaliar a atividade do tipo antidepressiva, mas não foi observado efeito significativo nas doses testadas. O maracujá alho potencializou o sono induzido por éter etílico, mostrando uma atividade hipnótico-sedativa. No teste de convulsão induzida por pentilenotetrazol, observou-se que o maracujá alho (400 mg/kg) exerceu um efeito protetor, diminuindo a severidade das convulsões e não causando a morte dos animais. Na avaliação do efeito anti-parkinsoniano, os testes comportamentais indicaram que o consumo de maracujá alho causou uma melhoria e possível efeito de recuperação dos danos induzidos pela rotenona no SNC, e além disso, provocou um aumento no nível de dopamina no cérebro dos animais na dose de 400 mg/kg. Pode-se concluir que o maracujá alho não apresentou toxicidade, sendo seguro para o consumo humano, e apresentou efeito funcional promissor como agente ansiolítico, hipnótico-sedativo, anticonvulsivante e antiparkinsoniano, possivelmente relacionado com os compostos fenólicos presentes
