Structural determination of a proinflammatory lectin of Vaitarea guianensis (Aublet)

The most studied lectins are of vegetable origin, mainly of the family Leguminosae. Within this family stand out the tribes Phaseoleae, Vicieae and Dalbergieae belonging to the subfamily Papilionoideae. Among the lectins of the Dalbergieae tribe, we can highlight a lectin with binding affinity to N-acetyl-D-galactosamine / D-galactose extracted from Vatairea guianensis (VGL) seeds that presented vasodilator effect in vitro with mechanism involving nitric oxide. Some lectins from this same tribe are already characterized biochemically, but only the lectins of Pterocarpus angolensis, Centrolobium tomentosum, Platypodium elegans, Arachis hypogaea and Vatairea macrocarpa have their structures. Structural studies of lectins are used as tools to understand how these proteins behave in the various physiological processes. For lectins that do not yet have their crystallographic structures resolved, Bioinformatics is highlighted using methodologies that predict the three-dimensional structure of these proteins in complex with their ligands. Considering that structural studies with these proteins are still few, the present work had the objective to predict the structure, to study the affinity and the interactions of the lectin of Vatairea guianensis seeds in complex with different glycans applying techniques of bioinformatics, like homology modeling and docking Molecular, in addition to investigating its inflammatory effect in vivo. The VGL model was obtained by homology with Vatairea macrocarpa (VML) lectin and demonstrated common characteristics of legume lectins and the analysis of protein-linker interaction revealed favorable interactions with N-acetyl-D-galactosamine, D-galactose and sugars. As well as various biologically relevant N- and O-glycans. The in vivo paw edema test revealed that VGL induces the edematogenic effect involving prostaglandins, interleukins and the lectin carbohydrate recognition domain. Together, these data corroborate with work already reported in which VGL interacts with N-or O-glycans of target molecules, particularly those with galactosides in their structure, contributing to the inflammatory effect of the lectin.Amplamente distribuídas na natureza, as lectinas compreendem um grupo heterogêneo de proteínas de origem não imune, que possuem pelo menos um domínio não catalítico capaz de seletivamente reconhecer carboidratos, e interagir de maneira reversível a açúcares livres ou glicanos presentes em glicoproteínas ou glicolipídeos, sem alterar suas estruturas. As lectinas mais estudadas são de origem vegetal, principalmente da família Leguminosae. Dentro desta família destacam-se as tribos Phaseoleae, Vicieae e Dalbergieae pertencentes à subfamília Papilionoideae. Dentre as lectinas da tribo Dalbergieae, podemos destacar uma lectina com afinidade de ligação a N-acetil-D-galactosamina/D-galactose extraída de sementes de Vatairea guianensis (VGL) que apresentou efeito vasodilatador in vitro com mecanismo envolvendo óxido nítrico. Algumas lectinas desta mesmo tribo já estão caracterizadas bioquimicamente, mas apenas as lectinas de Pterocarpus angolensis, Centrolobium tomentosum, Platypodium elegans, Arachis hypogaea e Vatairea macrocarpa têm suas estruturas. Os estudos estruturais de lectinas são utilizados como ferramentas para compreender de que forma essas proteínas se comportam nos vários processos fisiológicos. Para as lectinas que ainda não possuem suas estruturas cristalográficas resolvidas, a Bioinformática ganha destaque utilizando metodologias que realizam uma predição da estrutura tridimensional destas proteínas em complexo com os seus ligantes. Considerando que estudos estruturais com estas proteínas ainda são poucos, o presente trabalho teve por objetivo predizer a estrutura, estudar a afinidade e as interações da lectina de sementes de Vatairea guianensis em complexo com diferentes glicanos aplicando técnicas de bioinformática, como modelagem por homologia e docking molecular, além de investigar o seu efeito inflamatório in vivo. O modelo de VGL foi obtido por homologia com a lectina de Vatairea macrocarpa (VML) e demonstrou características comuns das lectinas de leguminosas e a análise da interação proteína-ligante revelou interações favoráveis com N-acetil-D-galactosamina, D-galactose e açúcares relacionados, bem como vários N- e O-glicanos, biologicamente relevantes. O teste in vivo edema de pata revelou que a VGL induz o efeito edematogênico envolvendo prostaglandinas, interleucinas e o domínio de reconhecimento a carboidratos da lectina. Conjuntamente, esses dados corroboram com trabalhos já reportados em que VGL interage com N- ou O-glicanos de moléculas alvo, particularmente nos que apresentam galactosídeos em sua estrutura, contribuindo para o efeito inflamatório da lectina
