Effect of cecropia pachystachya in inflammation and oxidative stress in naproxene induced gastric ulcers in mice.

Peptic ulcer (UP) is a gastrointestinal disease in which acute and chronic inflammation of the stomach occurs, as well as duodenum, which can lead to abdominal pain, nausea, vomiting and other symptoms. UP is multifactorial by Helicobacter pylori, use of non-steroidal anti-inflammatory drugs, alcohol, smoking and cancer treatment, resulting in an imbalance between the defensive and harmful factors. Cecropia pachystachya (CP) contains flavonoids, terpenes and polyphenols, such as: the chlorogenic acid known for its antioxidant properties. The present project investigated how explanations and antiulcerogenic properties of the leaf extract ethanolic extract of CP in gastric ulcer induced by naproxen (NPX). Male Swiss mice were pretreated with carboxymethylcellulose (0.5% vc CMC) and / or CP [1], [3], [10] and [30] mg / kg. After 30 minutes, they received NPX (300 mg / kg) animals were euthanized 6 hours after the experimental protocol. Microscopes, GSH, MDA, MPO, nitrite / nitrate, TNF-a and IL-6. The stomach segments were collected for analysis. Intestinal epithelial cells (IEC-6) were used to measure cell viability by the MTT test. The macroscopic test was performed by gas evaluation planning, a microscopy using a scoring system to define how edema, loss of cell architecture, hemorrhage and cell infiltration. Tests such as GSH and MDA were performed to evaluate oxidative stress. MPO, IL-6 and TNF-α analyzed CP antiinflammatory products. The MTT result showed that the CP extract was not different from the control with the culture medium in IEC-6. CP extract significantly reduced the macroscopic and microscopic gastric lesion induced by NPX. Cecropia pachystachya at 3.0 mg / kg prevented depletion at GSH concentrations, reduced as MDA concentrations and reduced neutrophil infiltration. The extract Cecropia pachystachya reduced as concentrations of the proinflammatory cytokines TNF-α and IL-6 and reversed the effect of NPX on nitrite / nitrate. On the other hand, CP demonstrates an anti-inflammatory and antioxidant effect without a model of gastric ulcer induced by NPX. This is the first work that shows the gastroprotection of this natural product.A úlcera péptica (UP) é uma doença gastrintestinal na qual ocorre inflamação aguda e crônica do estômago, assim como, duodeno, o que pode acarretar dor abdominal, náuseas, vômito entre outros sintomas. A UP é multifatorial por Helicobacter pylori, uso de antiinflamatórios não esteroidais, álcool, fumo e tratamento para o câncer, resultando de um desequilíbrio entre os fatores defensivos e nocivos. A Cecropia pachystachya (CP) contém flavonóides, terpenos e polifenóis, como: o ácido clorogênico conhecido por suas propriedades antioxidantes. O presente projeto investigou as possíveis atividades e propriedades antiulcerogênicas do extrato etanólico de folhas da CP na úlcera gástrica induzida por naproxeno (NPX). Os camundongos Swiss machos receberam pré tratamento com carboximetilcelulose (CMC 0,5% v.o) e/ou CP [1], [3], [10] e [30] mg/kg. Após 30 minutos receberam NPX (300 mg/kg) os animais foram eutanásiados 6 horas após o protocolo experimental. Os segmentos de estômago foram coletados para a análise dos seguintes parâmetros: macroscópia, microscópia, GSH, MDA, MPO, nitrito/nitrato, TNF-α e IL-6. Células epiteliais intestinais (IEC-6) foram utilizadas para estudar a viabilidade celular pelo teste do MTT. O teste da macroscopia foi realizado por planimetria para avaliar a lesão gástrica, a microscopia utilizou um sistema de escores para definir parâmetros como edema, perda de arquitertura das células, hemorragia e infiltrado de células. Testes como GSH e MDA foram realizados para avaliação do estresse oxidativo. MPO, IL-6 e TNF-α analisaram os efeitos antiinflamatórios da CP. O resultado do MTT mostrou que o extrato de CP não foi diferente do controle com meio de cultura em IEC-6. O extrato CP reduziu significativamente a lesão gástrica macroscópica e microscópica induzida pelo NPX. A Cecropia pachystachya na dose 3,0 mg/kg preveniu a depleção nas concentrações de GSH, reduziu as concentrações de MDA e reduziu a infiltração neutrofilica. O extrato Cecropia pachystachya reduziu as concentrações das citocinas pró-inflamatórias TNF-α e IL-6 e reverteu o efeito do NPX sobre nitrito/nitrato. Portanto, a CP demonstrou ter um efeito antiinflamatório e antioxidante no modelo de úlcera gástrica induzida por NPX. Este é o primeiro trabalho que mostra a gastroproteção deste produto natural
