Phytochemical study bioguided by the myorelaxant activity of the crude extract from stem bark of Hymenaea courbaril L (Jatobá)

Hymenaea courbaril L. (Fabaceae), popularly known as "Jatoba", is used in folk medicine to treat respiratory diseases such as bronchitis and asthma. This study evaluated the effect of H. courbaril on the contractile of rat isolated trachea, as well as its potential antioxidant and anti-inflammatory, through an interdisciplinary study bioguided by myorelaxant activity of the ethanol extract from stem bark (EEHC). Chromatoghrafic fractionation of EEHC resulted in fractions of increasing polarity: hexane (HF), hexane:dichloromethane (HDF), dichloromethane (DF), dichloromethane:ethyl acetate (DEAF), ethyl acetate (EAF) and methanol (MF) fraction. EEHC, DF, DEAF, EAF and MF showed enhanced free radical scavenging in the DPPH test. EEHC, DF, DEAF and MF partially relaxed tracheal rings contracted with either CCh or K+, while EAF almost abolished such contractions. EAF was more potent to inhibit K+ than CCh-induced contractions. EAF reduced contractions that depended on divalent cation inflow through voltage-operated Ca2+ channels (VOCCs) or receptor-operated Ca2+ channels (ROCCs), but it was more potent to inhibit VOCC- than ROCC-dependent contraction in ACh-enriched Ca2+-free medium, indicating a preferential action in events mediated electromechanically. Oral pre-treatment of antigen-challenged animals with EAF prevented airways hyperresponsiveness on KCl-induced contractions and impaired the increase in the number of white cells, particularly eosinophils and neutrophils in bronchoalveolar lavage of these animals. Chemical investigation of the most bioactive fraction (EAF) was performed using chromatographic methods and 1H-NMR and 13C-NMR spectroscopic analysis, resulting in the isolation and identification of the flavonoid astilbin and oleanolic acid, a compound triterpenoid, which is isolated by the first time in Hymenaea. Astilbin showed myorelaxant activity on rat trachea rings contracted with K+, indicating that it may be partly responsible for the activity presented by EAF. This study provided scientific basis that H. courbaril presents potential antioxidant, anti-inflammatory and myorelaxant, which support its use in folk medicine to treat inflammatory airway diseases such as asthma.Hymenaea courbaril L. O presente trabalho teve como objetivo investigar o efeito de H. courbaril na contratilidade da musculatura lisa traqueal de ratos, bem como seu potencial antioxidante e anti-inflamatório, através de um estudo farmacoquímico bioguiado pela atividade miorrelaxante do extrato etanólico das cascas do caule (EEHC). O tratamento cromatográfico de EEHC resultou em frações de polaridade crescente denominadas hexano (FH), hexano:diclorometano (FHD), diclorometano (FD), diclorometano:acetato de etila (FDAE), acetato de etila (FAE) e metanol (FM). EEHC, FD, FDAE, FAE e FM apresentaram potencial antioxidante através da atividade sequestradora de radicais livres, determinado pelo teste do DPPH. EEHC, FD, FDAE e FM relaxaram parcialmente anéis de traqueia contraídos com CCh ou K+, enquanto FAE se mostrou mais bioativa, revertendo completamente as contrações induzidas por esses agentes. FAE reduziu contrações que dependiam do influxo de cátions bivalentes através de canais de Ca2+ operados por voltagem (VOCs) ou canais de Ca2+ operados por receptor (ROCs), sendo mais potente em inibir as contrações dependentes de VOC, em meio sem Ca2+ e na presença de acetilcolina, evidenciando sua ação inibitória preferencial em eventos iniciados eletromecanicamente. O pré-tratamento de animais com FAE por via oral preveniu a hiperresponsividade das vias aéreas às contrações induzidas por KCl em traqueias de animais desafiados à ovalbumina, bem como reduziu a quantidade de células brancas, em especial, eosinófilos e neutrófilos no lavado broncoalveolar de animais sob processo asmático. A prospecção dos constituintes químicos da fração mais bioativa (FAE) foi realizada utilizando métodos cromatográficos e obtenção de espectros de RMN de 1H e 13C, resultando no isolamento e caracterização do flavonóide astilbina e do ácido oleanólico, um composto triterpenóide isolado pela primeira vez do gênero Hymenaea. Astilbina apresentou atividade relaxante sobre músculo liso traqueal de ratos pré-contraídos com K+, sendo responsável, pelo menos em parte, pela atividade apresentada por FAE. O presente estudo forneceu base científica de que H. courbaril apresenta potenciais atividades antioxidante, miorrelaxante e anti-inflamatória, respaldando o seu uso na medicina popular para o tratamento de doenças inflamatórias das vias aéreas, tais como a asma
