Estudo fitoquímico e abordagem sobre a atividade larvicida de Helicteres Eichleri K. Schum (Sterculiaceae) frente a Aedes aegypti L. (Diptera: Culicidae)

Since the dawn of human existence, populations around the world have been using natural products for medicinal purposes and spreading this knowledge for generations. In contemporary times, with the advances of science, the interest of researchers who seek in the medicinal plants the discovery of new bioactive substances has been intensified. In the world scenario, Brazil stands out as one of the countries with the greatest prospects for the sustainable exploitation of its biodiversity, since it holds the greatest diversity of plant species in the world. In this context, we emphasize the importance of research on plant species that present themselves as natural sources to combat the various stages of the Aedes aegypti mosquito, the main vector responsible for the transmission of dengue, zika, chikungunya and yellow fever viruses. This search for natural insecticides from renewable resources that are rapidly degradable, less damaging to the environment and human health, and the study of the use of Helicteres velutina in indigenous medicine against insect bites have led us to isolate and identify the chemical constituents, as well as to evaluate the larvicidal potential against Aedes aegypti of the species Helicteres eichleri K. Schum (Sterculiaceae), popularly known in Brazil as "monkey smoke".Through a chemical and biological pioneer study of the crude ethanolic extract obtained from the aerial parts of this species and using chromatographic and spectroscopic techniques, substances were isolated and identified, respectively: decanol (He-1), rosmarinic acid (He -2), a mixture of βsitosterol and stigmasterol (He-3a and He-3b), lupeol (He-4), a mixture of 3-α-hydroxy-urs-12en-28-oic and 3-α-hydroxy-olean-12-en-28-oic (He-5a and He-5b), a mixture of micromeric acid and ursolic acid (He-6a and He-6b). A preliminary approach, based on the exposure of the larvae in the L4 stage of Aedes aegypti to different concentrations of the crude ethanolic extract from the aerial parts of Helicteres eichleri, demonstrated promising larvicidal activity. After 24 and 48 hours of exposure, the concentration of 20mg/mL was able to kill 92.5% and 93.3% of the larvae, respectively. The LC50 was calculated at 4.4 mg/mL for the 24-hour interval, and, using the Tukey's test model, after 48 hours of exposure, LC50 was 3.73 mg/mL, thus characterizing a favorable alternative to be used in an integrated control system of this vector.Coordenação de Aperfeiçoamento de Pessoal de Nível Superior - CAPESDesde os primórdios da existência humana, populações de todo o mundo vem utilizando produtos naturais com fins medicinais e difundindo esses conhecimentos por gerações. Na contemporaneidade, com os avanços da ciência, intensifica-se o interesse de pesquisadores que buscam nas plantas medicinais a descoberta de novas substâncias bioativas. No cenário mundial, o Brasil se destaca como um dos países com maiores perspectivas para exploração sustentável de sua biodiversidade, uma vez que é detentor da maior diversidade de espécies vegetais do mundo. Nesse contexto, a importância das pesquisas sobre espécies vegetais que se apresentam como fontes naturais de combate aos vários estágios do mosquito Aedes aegypti, principal vetor responsável pela transmissão dos vírus da dengue, zika, chikungunya e febre amarela. Essa busca por inseticidas naturais, oriundos de recursos renováveis, rapidamente degradáveis, menos prejudiciais para o meio ambiente e para a saúde humana, e o estudo do uso da espécie Helicteres velutina na medicina indígena contra picadas de insetos, levou-nos a isolar e identificar os constituintes químicos, como também avaliar o potencial larvicida contra o Aedes aegypti da espécie Helicteres eichleri K. Schum (Sterculiaceae), conhecida popularmente no Brasil como “fumo-de-macaco”. Através de um estudo pioneiro do ponto de vista fitoquímico e biológico do extrato etanólico bruto obtido das partes aéreas desta espécie e utilizando técnicas cromatográficas e espectroscópicas, foram isoladas e identificadas, respectivamente, as substâncias: decanol (He-1), ácido rosmarínico (He-2), uma mistura do βsitosterol e estigmasterol (He-3a e He-3b), lupeol (He-4), uma mistura dos ácidos 3-α-hidróxiurs-12-en-28-óico e 3-α-hidroxi-olean-12-en-28-óico (He-5a e He-5b) e uma mistura dos ácidos micromérico e ursólico (He-6a e He-6b). Uma abordagem preliminar, a partir da exposição das larvas no estágio L4 de Aedes aegypti a diferentes concentrações do extrato etanólico bruto das partes aéreas de Helicteres eichleri, demonstrou promissora atividade larvicida. Segundo as análises estatísticas, em 24 e 48 horas de exposição, a concentração de 20mg/mL foi capaz de matar 92,5% e 93,3% das larvas, respectivamente. A CL50 foi calculada em 4,4 mg/mL para o intervalo de 24 horas e após 48 horas registrou uma CL50 de 3,73 mg/mL, caracterizando-se como uma alternativa favorável a ser utilizada em um sistema de controle integrado desse vetor
