Detecção de Leishmania (Ross, 1903) em Rattus norvegicus (Berkenhout,1769) em Belo Horizonte, Minas Gerais, 2007 e 2008

O objetivo da pesquisa foi o rastreamento por isolamento e cultura em meio bifásico, aposição de tecidos em lâminas, ensaio imunoenzimático (ELISA), imunofluorescência indireta (IFI) e LnPCR seguida de seqüenciamento de Leishmania sp. em Rattus norvegicus capturados em área de transmissão de leishmaniose em Belo Horizonte, Minas Gerais. As técnicas de diagnóstico direto (isolamento e cultura) e lâminas não revelaram formas promastigotas e amastigotas visíveis. Os testes sorológicos identificaram 43,75% (35/80) animais positivos no ELISA. Na IFI cada amostra foi pesquisada nas diluições 1:40, 1:80 e 1:160, onde 70% (56/80) dos animais foram positivos (10% a 1:160; 33,75% a 1:80 e 26,25% a 1:40) e 30% (24/80) negativos. Um total de 21,25% (17/80) dos animais foi negativo em ambos os testes. A nested PCR (LnPCR) dirigida ao gene SSUrRNA para Leishmania associada ao sequenciamento foi utilizada para analisar 315 amostras clínicas de 80 Rattus norvegicus. Amostras de 80 fragmentos de pele de cauda, 78 fragmentos de medula óssea, 78 amostras de sangue e 79 fragmentos de baço foram submetidos ao diagnóstico e revelaram 17,46% (55/315) tecidos positivos onde 10% (8/80) pele de cauda, 26,92% (21/78) sangue, 30,76% (24/78) medula óssea e 2,53% (2/79) baço. A taxa de infecção dos roedores foi 36.25% (29/80) e em 65.51% (19/29) destes os parasitos foram caracterizados como pertencentes ao complexo Leishmania braziliensis. Os resultados demonstram a urbanização do ciclo da leishmaniose tegumentar, tipicamente rural e adaptabilidade do vetor a ambientes antropizados. A identificação de infecção natural de R.norvegicus por L. braziliensis é importante indicador do potencial zoonótico desta espécie e a manutenção dos programas de controle populacional de roedores e de educação em saúde são medidas importantes no controle das leishmanioses no meio urbano, onde o contexto ambiental é constantemente modificadoThe cutaneous leishmaniasis (CL) is widespread and involves many species of Leishmania and a variety of vectors and hosts in a complex epidemiology. Diagnostic techniques (direct isolation and culture) revealed no visible amastigotes and promastigotes in samples analysed. Serological tests identified 43.75% (35/80) positive animals by ELISA. The infection rate of rodents was 36.25% (29/80) and 65.51% (19/29) of them had the parasite characterized as belonging to the Leishmania braziliensis. The results demonstrate the urbanization of the cycle of cutaneous leishmaniasis, typically rural and adaptability of the vector to the anthropogenic environments. The identification of natural infection by L. braziliensis in R. norvegicus is a significant indicator for zoonotic potential of this species and the maintenance of programs for its population control and health education are important measures in the control of leishmaniasis in the urban context where the environment is constantly modifie
