Testes moleculares como a PCR têm sido crescentemente utilizados em diagnósticos microbiológicos. Neste trabalho foi utilizado um protocolo de extração de DNA diretamente em amostras de leite e a utilização de uma PCR multiplex (mPCR) para identificação dos principais agentes da mastite bovina: Staphylococcus. aureus, Streptococcus agalactiae, Streptococcus uberis e Streptococcus dysgalactiae e do patógeno emergente: S. aureus resistente a meticilina (MRSA). Foram realizados testes com leite artificialmente contaminado com concentração bacteriana conhecida e posteriormente, para validar a técnica, foram analisadas 30 amostras oriundas de leite de tanque de expansão recebidas pelo Laboratório de Análise da Qualidade do Leite LabUFMG - MAPA. Para a realização das PCRs foram utilizados iniciadores que amplificam a região 16S-23S do RNA ribossomal de S. aureus, S. agalactiae, S. uberis e S. dysgalactiae e, para detecção de MRSA, utilizou-se iniciadores que amplificam o gene Mec A. O limite de detecção dos microrganismos envolvidos em infecções de glândula mamária é 1000 UFC/mL por mPCR, o que demonstra a sensibilidade do método para o diagnóstico etiológico de mastite bovinaEfficient control for mastitis requires sensitive, fast and specific tests to identify the agents that cause the disease.
