Antagonismo de bactérias da microbiota fecal humana contra enteropatógenos

A RC pode ocorrer pela produção de substâncias antagonistas e/ou pela competição por nutrientes e sítios de ligação. Para identificar as bactérias responsáveis e os mecanismos envolvidos na RC, tem-se utilizado o modelo animal isento de germes (IG), pois estudos in vitro nem sempre podem ser extrapolados para o que ocorre in vivo. No presente trabalho, após padronização da metodologia, ensaios de antagonismo ex vivo contra sete bactérias enteropatogênicas reveladoras, utilizando fezes de 15 humanos saudáveis, confirmaram que a RC tem variação individual, com sete voluntários (46%) apresentando uma alta RC (seis ou mais reveladoras inibidas), quatro voluntários (27%) com RC intermediária (quatro a cinco reveladoras inibidas) e quatro voluntários (27%) com baixa RC (uma a duas reveladoras inibidas). Utilizando ensaios de antagonismo in vitro e com sobrenadante de cultura, 14 bactérias isoladas da microbiota fecal dominante dos voluntários com boa RC foram selecionadas para associação em animais IG. No teste de antagonismo in vivo contra Salmonella enterica sor. Typhimurium, camundongos monoassociados com as linhagens Enterococcus hirae (8.2), Bacteroides thetaiotaomicron (16.2) e Lactobacillus ruminis (18.1) apresentaram reduções significativas nas contagens fecais do patógeno ao longo do desafio. Após cinco dias de infecção, no grupo 8.2, observou-se redução (p < 0,05) na translocação de S. Typhimurium para o baço, enquanto no grupo 18.1 houve aumento (p < 0,05) da translocação para o fígado. Esses resultados foram confirmados pelas avaliações histopatológicas, em que apenas o grupo 8.2 mostrou evidentes sinais de proteção no íleo e no fígado, em relação aos danos causados pelo desafio com S. Typhimurium num grupo controle, enquanto que no grupo 18.1 houve lesões significativamente mais intensas. Concluindo, a partir a microbiota fecal dominante de doadores humanos saudáveis apresentando uma elevada RC e utilizando ensaios ex vivo, in vitro e in vivo, foi possível selecionar uma bactéria com um potencial alto de RC.The term "indigenous microbiota" refers to the population of micro-organisms normally present on the surface and mucosa of an individual, where performs functions essential to the health of the host, including the colonization resistance (CR) by pathogens. The CR may occur due to the production of antagonistic substances and/or the competition for nutrients and binding sites. To identify the bacteria responsible and the mechanisms involved in the CR, the germ-free (GF) animal model has been used, because in vitro studies cannot always be extrapolated to what occurs in vivo. In the present study, after standardization of the methodology, ex vivo antagonism tests against seven enteropathogenic indicator bacteria using feces from 15 healthy human volunteers, confirmed that the CR showed individual variation, with seven volunteers (46%) having a high CR (six or more indicators inhibited), four subjects (27%) an intermediate CR (four to five indicators inhibited), and four subjects (27%) a low CR (one to two indicators inhibited). Using in vitro antagonism assays and with culture supernatant, 14 strains isolated from fecal dominant microbiota of volunteers with elevated CR were selected for association in GF mice. In the in vivo antagonism assay against Salmonella enterica ser. Typhimurium, mice monoassociated with Enterococcus hirae (8.2), Bacteroides thetaiotaomicron (16.2) and Lactobacillus ruminis (18.1) strains had significant reductions in fecal counts of the pathogen during the challenge. After five days of infection, the 8.2 group showed a reduction (p < 0.05) in the translocation of S. Typhimurium to the spleen, while the 18.1 group had an increased (p < 0.05) translocation to the liver. These results were confirmed by the histological data, in which only the 8.2 group showed clear protective signs of ileum and liver, compared to the damage caused by challenge with S. Typhimurium in a control group, while in group 18.1 there was significantly more intense lesions.
