Avaliação da atividade de telomerase em células-tronco embrionáriasindiferenciadas e em células neoplásicas de camundongo

Sua função principal é reconstruir os telômeros, seqüências repetitivas situadas nas pontas de cada cromossomo, que protegem contra danos no ácido desoxirribonucléico (DNA) e que também funcionam como relógio biológico, limitando o número de vezes pelas quais a célula pode se dividir. A telomerase encontra-se ativa em células com alta capacidade de replicação, como célulastronco e células neoplásicas, e não é detectada na maioria das célulasdiferenciadas de um organismo adulto. O estudo da telomerase pode resultar em maneiras de se controlar, aumentando-se ou inibindo-se, a capacidade de replicação celular, situação bastante útil tanto no campo da oncologia quanto no processo de terapia celular com células-tronco. Os objetivos deste trabalho foram: avaliar a atividade da telomerase (AT) em diferentes passagens de células-tronco embrionárias indiferenciadas de camundongo (CTEc) e comparar a AT das CTEccom linhagens de células de tumor de camundongo. Foi utilizado o método TRAPELISA para avaliação semiquantitativa da AT. Foram estudadas três linhagens de CTEc, denominadas CT2, CT3 e CT4. As passagens celulares avaliadas foram as de número 13, 15 e 19, denominadas P13, P15 e P19, respectivamente. Todas as passagens celulares foram avaliadas em relação ao estado de indiferenciação, apartir da presença dos genes OCT4 e NANOG. As linhagens de tumor avaliadas foram B16F1 (melanoma), B16F10 (melanoma metastático) e CT26WT (tumor de cólon). Os resultados são expressos em porcentagem diante de um padrão com alta AT conhecida. A CT2 apresentou 1,09% em P13, 13,24% em P15 e 36,90% em P19. A CT3 apresentou 1,46% em P13, 21,82% em P15 e 22,37% em P19. ACT4 apresentou 3,82% em P13, 39,05% em P15 e 34,22% em P19. A média das três linhagens foi P13 1,59%, P15 18,53% e P19 23,37%. As linhagens de tumor B16F1, B16F10 e CT26WT apresentaram, respectivamente, 128,60, 1,45 e 30,94%. Nas três passagens de CTEc avaliadas, observou-se tendência a aumento inicial, com posterior estabilização da AT. As linhagens celulares de tumor de camundongo apresentam AT com valores mais variáveis. Os resultados permitem concluir que tanto as CTEc quanto as linhagens de tumor apresentamalta capacidade de multiplicação, expressa pelas medidas elevadas de AT encontradas.Telomerase is an enzyme which plays a role in the maintenance of theproliferation ability of the cells and in the protection and stabilization of thechromosomes. The main function of telomerase is to add telomere sequences at the end of each chromosome, protecting it from DNA damage, and functioning as a biological clock, limiting the maximum number of divisions each cell can undergo. The telomerase activity (TA) can be detected in highly replicative cells, like stem cells and cancer cells, but not in the majority of the differentiated cells of the human organism. The study of telomerase can result in efficient forms ofcontrolling the cell proliferation ability, can also be useful both in the treatment of cancer and also in the researches concerning stem cell therapy. The purpose of this study is to evaluate the TA in different passages of mouse embryonic stem cells (mESC) and to compare the TA of the mESC with mouse neoplastic cells. TRAP-ELISA method was used in a semiquantitative evaluation of TA. Three mESC lineages were evaluated, named CT2, CT3 and CT4 respectively, in three different passages (P13, P15 and P19) each one. All the samples were evaluatedaccording to the presence of undifferentiation markers (genes OCT4 and NANOG) All the samples were evaluated according to the presence of undifferentiation markers (genes OCT4 and NANOG). The cancer lineages B16F1, B16F10 e CT26WT showed 128.60%, 1.45% and 30.94% respectively. The results obtained demonstrated allthe samples with high TA. The three passages of mESC showed a tendency of increase in TA and subsequent stabilization. The cancer lineages showed more variable TA values. The results achieved after data analysis let us conclude that both mESC and cancer lineages demonstrate a high capacity of proliferation, due to the high values of TA found in this study
