Diversidade genética em taperebazeiro (Spondias mombin L., Anacardiaceae)

It presents economic and social importance and it is widely used by the industry for making juices, popsicles, ice cream, jams and liqueurs. This study aimed to characterize the genetic diversity within and among populations of S. mombin. The analysis was performed with molecular markers AFLP (Amplified Fragment Length Polymorphism) in three populations located in the city of Manaus, Amazonas State: Federal University of Amazonas (UFAM), Vila Buriti (VLB) and the Executive Committee of the Cocoa crop Plan (CEPLAC). 10 combinations of oligonucleotides were tested, and four selected for this analysis: E + AAC / M + CAT, E + AGT / M + CTC, E + AAC / M + CTC, E + AGT / M + CAT. The oligonucleotides revealed 283 polymorphic loci, ranging from 97.2% to 63% among populations. The Fst (genetic differentiation between populations) figured was 0.52, showing a high level of genetic differentiation among the populations. The result of Analysis of Molecular Variance attributed 52.8% and 47.1% of the variation among and within populations, respectively. The dendrogram based on AFLP markers revealed the formation of two groups, the natural, UFAM and VLB, and CEPLAC group, which is a population enriched with planting. The AFLP markers are efficient to detect genetic diversity in S. mombin and differentiate populations of different constitutions. Most of the genetic variability occurs between populations.O taperebazeiro (Spondias mombin L.) é uma fruteira pertencente à família Anacardiaceae e tem uma distribuição geográfica ampla. Apresenta importância econômica e social e é bastante utilizado pela indústria para confecção de sucos, picolés, sorvetes, geleias e licores. Este estudo teve como objetivo principal caracterizar a diversidade genética entre e dentro de populações de S. mombin L. A análise foi realizada por meio de marcadores moleculares AFLP (Amplified Fragment Length Polymorphism) em três populações localizadas na cidade de Manaus, Estado do Amazonas: Universidade Federal do Amazonas (UFAM), Vila Buriti (VLB) e Comissão Executiva do Plano da Lavoura Cacaueira (CEPLAC). Foram testadas 10 combinações de oligonucleotídios, e selecionadas quatro para essa análise: E+AAC/M+CAT, E+AGT/M+CTC, E+AAC/M+CTC, E+AGT/M+CAT. Os oligonucleotídios revelaram 283 locos polimórficos, variando de 63% a 97,2% entre as populações. O Fst (diferenciação genética entre as populações) calculado foi de 0,52, revelando um alto nível de diferenciação genética entre as populações. O resultado da Análise Molecular de Variância atribuiu 52,8% e 47,1% da variação entre e dentro das populações, respectivamente. O dendrograma construído com base nos marcadores AFLP revelou a formação de dois grupos, o das populações naturais, UFAM e VLB, e o grupo da CEPLAC, que é de uma população enriquecida com plantio. Os marcadores moleculares AFLP são eficientes para detectar diversidade genética em S. mombin e diferenciar populações de constituições distintas.
