Desenvolvimento de extrato seco padronizado de frutos sem sementes de Caesalpinia ferrea C. Mart. e avaliação da atividade anti-inflamatória

Mart (Fabaceae), known in Amazonian region as jucá is widely used in the folk medicine in inflammation and to treat wounds and anemia. However, there are no studies on the quality control of the C. ferrea raw material, lacking the standardization of the chemical and extractive procedures, identifying the substances responsible for the pharmacological effects. In this work, after the raw material characterization, two standardized extracts were produced and their analgesic/anti-inflammatory activities were compared. The aqueous extracts were obtained by decoction of the fruits without seeds at 2,5%. After filtration and concentration, the extracts were dried by aspersion or freeze-dryer, generating the ADE - aspersion dried extract, and the FDE freeze-dried extract. ADE and FDE were standardized by the humidity ratio and total tannins and polyphenols concentrations. The humidity ratio (14% and 11%), total tannins (13% and 11%) and polyphenols (49% and 43%) were not different between ADE and FDE, respectively. In the general pharmacological test, the standardized extracts (0,1; 0,3 e 1,0 g/kg, p.o.), did not cause ataxia, motor incoordination, irritability or hiper-reactivity to nociceptive stimulation. Death was not observed, showing that the extracts did not produce potential toxic effects. At the major doses, the extracts increased the ethyl ether and the pentobarbital induced-sleeping time, indicating the depression of the central nervous system without affecting the motor activity, as confirmed in the rota-rod test. In the formalin test, the extracts reduced the lickingtime in the 1st and in the 2nd phase; this effect was blocked by the previous (15 min) administration of naloxone (5 mg/kg, i.p.). In the acetic acid-induced abdominal contortions test, the extracts (0,1; 0,3 e 1,0 g/kg, p.o.) diminished the number of contortions until 30 min after acetic acid administration. The extracts neither change the basal latency of the thermal stimulation reaction (55 ºC) in the tail-flick test nor inhibited the carrageenan-induced acute paw edema; the positive control indomethacin inhibited the edema in 60% after 2 h. In the dextran-induced acute paw edema test, the treatment with ADE or FDE reduced the edema in 30%, without dose-effect relationship; the positive control cyproheptadine (10 mg/kg, p.o.) reduced the edema in 81% in the same time. With these results, we concluded that the pharmacological effects were not different between the two standardized extracts - ADE and FDE from Caesalpinia ferrea C. Mart. The anti-nociceptive effect in the extracts containing ~46% polyphenols and ~12% tannins involves common pathways to the opioids. ADE and FDE inhibited the dextran-induced acute paw edema, but did not inhibit the carrageenaninduced acute paw edema.Caesalpinia ferrea C. Mart (Fabaceae), conhecida na Amazônia como jucá, é amplamente utilizada na medicina popular como anti-inflamatória, cicatrizante e no tratamento da anemia. Entretanto, não há estudos que garantam a qualidade da matéria-prima de C. ferrea, sendo necessárias padronização de técnicas de extração e padronização química com identificação das substâncias responsáveis pelos efeitos farmacológicos. Neste trabalho, após caracterização da matéria-prima, foram padronizados dois extratos de C. ferrea e comparadas as atividades anti-inflamatória e analgésica dos mesmos. Os extratos aquosos foram obtidos por decocção dos frutos sem sementes a 2,5%. Após filtração e concentração, os extratos foram secos por aspersão ou liofilização, dando origem ao ESA extrato seco por aspersão e ao ESL extrato seco por liofilização. O ESA e ESL foram padronizados segundo o teor de umidade e concentrações de polifenóis e de taninos totais. O teor de umidade (14% e 11%), taninos totais (13% e 11%) e polifenóis (49% e 43%) não foram diferentes entre ESA e ESL, respectivamente. No teste farmacológico geral em camundongos, os extratos padronizados nas doses de 0,1; 0,3 e 1,0 g/kg, v.o., não produziram sinais de ataxia, incoordenação motora, irritabilidade ou hiperreatividade a estímulos nociceptivos. Não houve morte entre os animais, demonstrando que os extratos não produziram efeitos tóxicos graves. Nas maiores doses, os extratos produziram aumento da duração do sono induzido por barbitúrico e por éter etílico, indicando ação depressora do SNC sem afetar, no entanto, a atividade motora, como observado no teste do rota-rod . No teste da formalina (1,5%), os extratos ESA e ESL diminuíram o tempo de lambedura na 1ª e na 2ª fase do teste; esse efeito foi bloqueado com a administração prévia (15 min) de naloxona (5 mg/kg, i.p.). No teste de contorções abdominais induzida por ácido acético (0,8%), os extratos (0,1; 0,3 e 1,0 g/kg, v.o.) diminuíram o número de contorções até 30 min após a injeção de ácido acético. Os extratos não mudaram a latência basal de reação ao estímulo térmico (55 ºC) no teste de retirada da cauda ( tail flick ) e nem inibiram o edema de pata induzido por carragenina (1%); o controle positivo indometacina inibiu o edema em 60% após 2 h. No teste do edema de pata induzido por dextrana (0,1%), o tratamento com ESA ou ESL inibiu o edema em 30%, sem relação dose-efeito; o controle positivo ciproheptadina (10 mg/kg, v.o.) reduziu o edema em 81% no mesmo tempo. Com esses resultados, concluímos que os efeitos farmacológicos não foram diferentes entre os dois extratos - ESA e ESL - padronizados de Caesalpinia ferrea C. Mart. Esses extratos contendo ~46% de polifenóis e ~12% de taninos têm efeito antinociceptivo envolvendo vias comuns aos opióides e atividade inibidora do edema induzido por dextrana, mas não inibem o edema induzido por carragenina
