Com o intuito de ratificar estes dados, 201 cães do município da Serra foram investigados para LV, utilizando testes sorológicos, parasitológicos e moleculares. Deste total, 13% (26/201) e 5,97% (12/201) apresentaram respectivamente resultados positivos no teste ELISA in house e Kalazar Detect. Nenhum animal foi soropositivo para os testes RIFI e ELISA (Biomanguinhos). Dois cães soropositivos apresentaram cultura positiva para Leishmania (Leishmania) chagasi e quatro animais apresentaram PCR positivo para o gênero Leishmania. Além disso, um óbito humano por LV foi registrado em associação a dois casos caninos da doença (ambos com Kalazar Detect positivo e um com cultura positiva). A investigação epidemiológica mostrou que ambos os cães foram nascidos e criados na Serra e não possuíam história de viagem para áreas endêmicas para LV. O outro cão com cultura positiva foi capturado pelo Centro de Controle de Zoonoses da Serra a 550 metros da residência da paciente. Apesar da espécie Lutzomyia longipalpis não ter sido encontrada, outras espécies de flebotomíneos foram capturadas: L. edwardsi, L. tupynambai, L. cortelezzii, L. sordellii e L. intermedia. Nossos resultados sugerem fortemente a existência da LV canina autóctone no município da Serra e o primeiro caso autóctone humano neste município, embora não confirmado devido a paciente ser natural de uma área endêmica para a doença.Although the Metropolitan Region of Vitória, Espírito Santo state, is considered a free visceral leishmaniasis (VL) area, a recent canine soroepidemiological inquiry revealed that 7 out of 158 (4,4%) stray dogs were seropositive (Kalazar Detect) for VL, suggesting the existence of autochthonous VL in this region. In order to clarify these findings, 201 dogs from Serra municipality were screened for VL, using serological, parasitological and molecular tests. Of this total, 13% (26/201) and 5,97% (12/201) were positive for ELISA in house and Kalazar Detect tests, respectively. None of the dogs was positive for RIFI and ELISA assays (Biomanguinhos). Two seropositive dogs presented positive culture for Leishmania (Leishmania) chagasi, and four were PCR positive for the Leishmania genus. Additionally, one human VL case was reported in association with 2 canine cases (both of them positive for Kalazar Detect and one had a positive culture). The epidemiological investigation showed that both dogs were born in Serra and never travelled to VL endemic areas. The other dog with a positive culture was captured by the Zoonosis Control Center 550 meters far from the patient s dwellings. Although the Lutzomyia longipalpis species was not detected, others phlebotomine sand fly species were found such as L. edwardsi, L. tupynambai, L. cortelezzii, L. sordellii, and L. intermedia. Our results strongly suggest the existence of autochthonous canine VL and the first autochthonous human case of the disease in Serra, though this cannot be confirmed because the patient was native from an endemic area for VL
