Reactions of trihalomethylsubstituted enones with amines and diamines. Synthesis of ethambutol analogues

Conselho Nacional de Desenvolvimento Científico e TecnológicoA series of 5,5,5-trifluoro-3-methyl-(alkylmethylamino)-pentane-1,4-diols, 5,5,5-trifluoro-3-methyl-(arylmethylamino)-pentane-1,4-diols and N,N-bis[5,5,5- trifluoro-3-methyl-(alkylmethylamino)]-pentane-1,4-diols were obtained from the reaction of 3-trifluoroacetyl-4,5-dihydrofuran with amines and diamines and subsequent reduction reaction using sodium borohydride (NaBH4) as a reducing agent. For the synthesis of amino alcohols derived from aryl amines, an equimolar amount of triethylamine was used in the first reaction step, providing better yields. The amino alcohols were obtained as diastereoisomeric mixtures, in the form of oil and with yields from moderate to good (50-83%). This work also presents the synthesis of β-dienamino ketones from the condensation of diamines with trihalomethylsubstituted enones using a simple and efficient method. The yields obtained were excellent and ranged from 75-98%. The trihalomethylsubstituted enones used as starting reagents were synthesized by acylation of acetals or enol ethers with trifluoroacetic anhydride or chloride trichoroacetil.Uma série de 5,5,5-trifluor-3-metil-(alquilmetilamino)-pentano-1,4-dióis, 5,5,5-trifluor-3-metil-(arilmetilamino)-pentano-1,4-dióis e N,N-bis[5,5,5-trifluor-3-metil-((alquilmetilamino]-pentano-1,4-dióis foram obtidos a partir da reação do 3-trifluoracetil-4,5-diidrofurano com aminas primárias e diaminas e subsequente reação de redução com borohidreto de sódio (NaBH4) como agente redutor. Para a síntese dos aminoálcoois derivados das aril aminas, uma quantidade equimolar de trietilamina foi utilizada na primeira etapa da reação, proporcionando melhores rendimentos. Os aminoálcoois foram obtidos como misturas diastereoisoméricas, em forma de óleo e com rendimentos de moderados a bons (50-83%). Este trabalho também apresenta a síntese de β-dienamino cetonas a partir da condensação de diaminas (1,2-etilenodiamina, 1,3-diaminopropano e 1,4-diaminobutano) com enonas trialometilsubstituídas utilizando uma metodologia simples e eficiente. Os rendimentos obtidos foram ótimos e variaram de 70-98%. As enonas trialometilsubstituídas usadas como reagentes de partida, foram sintetizadas a partir da acilação de acetais ou enol éteres com anidrido trifluoracético ou cloreto de tricloroacetila
