Identification of canine parvovirus in the wild animals Brazilian wildlife

O parvovírus canino (CPV) é o agente causal da parvovirose canina que ocasiona enterite e miocardite grave em cães jovens. O CPV tipo 2 (CPV-2) emergiu como uma variante do vírus da panleucopenia felina (FPV) o qual foi adaptado ao hospedeiro canino por carnívoros selvagens. Atualmente o CPV-2 é classificado em três subtipos: CPV-2a, CPV-2b e CPV-2c. Estes subtipos cocirculam em frequência variável dependendo da localidade geográfica. O cachorro-do-mato (Cerdocyon thous) está presente em todos os biomas brasileiros e em alguns países da América do Sul. O quati (Nasua nasua) pertence à fauna silvestre da América do Sul e está amplamente distribuído no território brasileiro. O presente estudo teve como objetivo verificar o envolvimento de agentes infecciosos virais como causa da morte de um filhote de cachorro-do-mato (Cerdocyon thous) e verificar a presença de vírus entéricos em uma população de quatis de vida livre. Amostras de fezes de 21 quatis (Nasua nasua) (ambos os sexos, diferentes faixas etárias e aspectos das fezes) foram coletadas de uma população de quatis que habita uma reserva ambiental em Palmital, São Paulo, para pesquisa de CPV-2 e Rotavírus (RV) por meio das técnicas de reação em cadeia da polimerase (PCR) e eletroforese em gel de poliacrilamida (EGPA), respectivamente. A PCR foi realizada com intuito de amplificar um fragmento de 583pb do gene VP2 do CPV-2. Para a pesquisa de agentes infecciosos virais, tais como o vírus da cinomose canina (CDV), o adenovírus canino A tipo 1 e tipo 2 (CAdV-1 e CAdV-2), o herpesvírus canino 1 (CaHV-1) e o CPV-2 como causa da morte de um filhote do cachorro-do-mato (Cerdocyon thous) de vida livre, fragmentos de órgãos foram coletados e submetidos a técnicas moleculares (PCR e RT-PCR). Os produtos de PCR do CPV-2 foram sequenciados e as sequências de nucleotídeo (nt) e aminoácido (aa) obtidas foram analisadas. Das 21 amostras de fezes de quatis analisadas, oito (38%) foram positivas para CPV-2 na técnica de PCR e nenhuma foi positiva para RV na EGPA. As amostras de quati positivas para CPV-2 exibiram 100% de identidade de nt com a cepa protótipo de CPV-2b (CPV-39) e na árvore filogenética agruparam com as cepas pertencentes ao CPV-2b. Dos oito fragmentos de órgãos do cachorro-do-mato avaliados apenas o fragmento do coração foi positivo para o CPV-2 na PCR e as amostras foram negativas para os outros vírus avaliados. A amostra de cachorro-do-mato positiva para CPV-2 apresentou 99,6% de identidade de nt com o protótipo CPV-2b (CPV-39) e na árvore filogenética agrupou com cepas CPV-2b. O presente estudo relata a detecção de CPV-2b em um filhote de cachorro-do-mato de vida livre no Brasil e também relata a presença de CPV-2b em quatis assintomáticos que podem eliminar o vírus nas fezes sendo importantes elos na transmissão e na permanência do CPV em populações de animais domésticos e silvestres.Canine parvovirus (CPV) is the causal agent of canine parvovirosis that causes enteritis and myocarditis in young dogs. The CPV type 2 (CPV-2) emerged as a variant of feline panleukopenia virus (FPV) which is adapted to the canine host by wild carnivores. Currently the CPV-2 is classified into three subtypes: CPV-2a, CPV-2b and CPV-2c. These subtypes co-circulate in a variable frequency depending of the geographic location. The Crab-eating fox (Cerdocyon thous) is present in all Brazilian biomes and in some countries of South America. The coati (Nasua nasua) belongs to the wildlife of South America and is widely distributed in Brazil. This study aimed to verify the involvement of viral infectious agents as the cause of death of a young Crab-eating fox (Cerdocyon thous) and to veirfy the presence of enteric viruses in a population of free-living coatis. Fecal samples of 21 coatis (Nasua nasua) (both sexes, different ages and aspects of faeces) were collected from a population of coatis that inhabits an environmental reserve in Palmital city, São Paulo, to search the CPV-2 and Rotavirus (RV) using the polymerase chain reaction (PCR) and polyacrylamide gel electrophoresis (PAGE), respectively. The PCR was performed to amplify a fragment of 583bp of the VP2 gene from CPV-2. For researching the viral infectious agents, such as canine adenovirus type 1 and type 2 (CADV-1 and CADV-2), canine herpesvirus type 1 (CaHV-1) and CPV-2 as cause of death of a puppy Crab-eating fox (Cerdocyon thous) free life, tissue fragments were collected and submitted to molecular techniques (PCR and RT- PCR). The CPV-2 PCR products were sequenced and the nucleotide (nt) and amino acid (aa) 25 sequences obtained were analyzed. Of the 21 fecal samples form coatis analyzed, 8 (38%) were positive for CPV-2 in PCR and none was positive to RVA in PAGE. The coati samples positive for CPV-2 exhibited 100% of nt identity with the CPV-2b prototype strain (CPV-39) and in the phylogenetic tree grouped with strains that belong to CPV-2b. Of the eight organ fragments of the Crab-eating fox evaluated only the heart fragment was positive for CPV-2 in PCR and the samples were negative for the other evaluated virus. The Crab-eating fox positive sample for CPV-2 showed 99.6% of nt identity with the CPV-2b prototype strain (CPV-39) and in the phylogenetic tree grouped with CPV-2b strains. This study reports the CPV-2b detection in a puppy Crab-eating fox free life in Brazil and also describes the presence of CPV-2b in asymptomatic coatis that can eliminate the virus in the feces and are important in the transmission and permanence of CPV in domestic and wild animal populations
