Postsynaptic role of SNARE proteins in synaptic plasticity

One of the most fascinating properties of the brain is synaptic plasticity, which may be defined as activi-ty-dependent modifications of structure and efficacy of synapses. Synaptic plasticity is believed to be the cellular basis of learning and memory. One of the mechanisms underlying synaptic plasticity is fusion of glutamate receptor-containing vesicles with the postsynaptic membrane. However, the underpinnings of postsynaptic vesicle exocytosis are poorly understood. SNAREs are proteins that mediate membrane fusion. This thesis has for the first time demonstrated ultrastructural localization of SNARE proteins in the postsynaptic spine. Furthermore, SNARE proteins were co-localized with AMPA- and NMDA-types of glutamate receptors. In addition, evidence in favour of SNARE-dependent trafficking of these recep-tors are provided. Moreover, this work has revealed altered synaptic SNARE concentrations in some neuropsychiatric diseases, which may possibly be linked to the pathogenesis of these disorders. Taken together, the results presented in this thesis will lead to a better understanding of the molecular mecha-nisms underlying synaptic plasticity in health and disease
