Abstrak. Kitin dapat diisolasi dari cangkang rajungan melalui tahap demineralisasi dan deproteinasi. Tujuan penelitian ini adalah untuk mengetahui perbandingan kitin dan enzim yang optimum dalam menghidrolisis protein pada kitin. Penentuan perbandingan kitin dan enzim dilakukan dengan cara memvariasaikan enzim dan kitin yang digunakan. Jumlah protein yang terhidrolisis ditentukan dengan menggunakan metode Kjeldahl. Kitin yang diperoleh berwarna putih. Perbandingan enzim dan kitin yang optimum adalah 1:10 (b/v) dengan kadar N-total sebesar 3,02%.
