This thesis concerns the identification of the genetic and environmental causes of complex diseases. Our central application is Isis-Diab a study of type 1 diabetes. We propose a method to use phase information to improve genetic risk predictions. We replicate an estimate of genetic risk on Isis-Diab patients. We prove an asymptotic equivalence result between two paired data analysis methods. We then analyze the Isis-Diab case-control study based on environmental questionnaires. We try to confirm the results of this study by crossing the genetic risk and environemental factors in Isis-Diab patients.
