When targeting neuroimaging of neuronal networks in live thick specimens with high temporal and spatial resolution, Light-Sheet Fluorescence Microscopy (LSFM) has demonstrated its capability to provide enhanced signal to background ratio while decreasing toxicity and photobleaching. However, imaging neuronal networks is still a challenge since fluorescence variations induced by neuronal activity are weak and imaging depth is limited because of optical aberrations. Adaptive optics has shown its ability to increase signal, resolution and imaging depth in LSFM through direct or indirect wavefront sensing but the latter are time consuming or impose complex implementation with a virtual guide-star.This thesis relates an original development of a...
