The effect of vitamin B complex therapy on the regeneration of motor function of the peripheral nerve in rat femoral nerve injury model

Povrede perifernih nerava predstavljaju problem velikih razmera u savremenom svetu. Posledice po radnu i ţivotnu aktivnost su velike usled ograniĉenih mogućnosti za regeneraciju, pogotovo motorne funkcije. Razliĉiti terapeutski pristupi su pokušali da poboljšaju rezultate motorne regeneracije. Znaĉajan broj studija je pokazao povoljan efekat razliĉitih vitamina B kompleksa na regeneraciju perifernog nerva (Liao i sar., 2010 i 2013; Fujii i sar., 1995; Talebi i sar.,2013; Turan i sar., 2014). U navedenim istraţivanjima su korišćeni razliĉiti eksperimentalni animalni modeli (Irintchev, 2011; Nichols i sar., 2005; Witehead i sar., 2005) kao i in vitro studije. U ovom radu je ispitivan uticaj hipervitaminske terapije vitaminima B kompleksa na regeneraciju motorne funkcije femoralnog nerva pacova. Dadatno je ispitivan i efekat povrede i primenjene terapije na peritonealne makrofage. Ciljevi: (I) Ispitati ćelijske mehanizme u regeneraciji i oporavku perifernog nerva nakon povrede i rekonstrukcije motorne grane n. femoralis-a; (II) Ispitati uticaj hipervitaminske terapije vitaminima B kompleksa na elektriĉnu aktivnost, atrofiju i funkciju ciljnog mišića; (III) Ispitati uticaj hipervitaminske terapije vitaminima B kompleksa na gustinu ćelijskih jedara u podruĉju povreĊenog nerva i proces neuroreparacije praćen ekspresijom specifiĉnog markera izrastanja aksona (GAP 43); (IV) Ispitati uticaj hipervitaminske terapije vitaminima B kompleksa na prisustvo satelitskih ćelija tokom oporavka funkcije mišića praćen ekspresijom specifiĉnog markera (Pax7); (V) Ispitati uticaj povrede motorne grane n. femoralis-a i primenjene hipervitaminske terapije vitaminima B kompleksa na funkciju peritonealnih makrofaga ukljuĉujući metaboliĉku vijabilnost, sposobnost fagocitoze, adhezije kao i produkcije reaktivnih oblika kiseonika, azot oksida (NO) i citokina. Materijal i metode: U eksperimentu je korišćeno 115 adultnih muţjaka Albino Oxford pacova teţine od 200 do 350 grama. Ţivotinje su podeljene na eksperimentalnu i kontrolnu grupu...Injuries of the peripheral nerves represent a high scale problem in modern world. They lead to significant consequences considering working ability and quality of life due to restricted recovery, especially of motor function. Different therapeutical approaches were used in order to improve motor recovery. Significant number of studies showed benefitial effect of different vitamins of B complex on motor regeneration (Liao, et al. 2010 and 2013; Fujii, et al., 1995; Talebi, et al.,2013; Turan, et al., 2014). Different experimental animal models were used in these researches (Irintchev, 2011; Nichols, et al., 2005; Witehead, et al., 2005) as well as in vitro studies. Macrophages, abundant and phenotypically diverse immune cell populations, play an important role during peripheral nerves regeneration . In this study we investigated effect of vitamin B complex therapy on regeneration of motor function of the peripheral nerve and characteristics of peritoneal macrophages (PMF) in rat femoral nerve injury model. Aims: (I) Investigate cell mechanisms involved in regeneration and recovery after injury and reconstruction of motor branch of femoral nerve; (II) Investigate effect of vitamin B complex therapy on electrical activitiy, atrophy and function of target musle; (III Investigate effect of vitamin B complex therapy on nuclear density in area of injured nerve and process of neuroreparation followed by expression of axon growth specific marker (Growth associated protein, GAP 43); (IV) Investigate effect of vitamin B complex therapy on presence of satellite cells during recovery phase of the muscle followed by expression of the specific marker (Paired box protein, Pax 7); (V) Investigate influence of femoral nerve motor branch injury and administered vitamin B complex therapy on function of PMF including metabolic viability, phagocytosis and adhesion abilities, as well as production of nitrogen oxide (NO), cytokines and reactive oxygen species (ROS).
