Biotechnological production of 2,3-butanediol by bacterium Paenibacillus polymyxa DSM 742

Povećanje potraţnje 2,3-butandiola potaknulo je istraţivanja biotehnološke proizvodnje 2,3-butandiola sa ciljem zamjene dosadašnje kemijske proizvodnje. U ovom radu istraţivana je proizvodnja 2,3-butandiola s bakterijom Paenibacillus polymyxa DSM 742, tijekom kojeg se ispitivao rast pri različitom pH (3, 4, 5 i 6,5), utjecaj broja okretaja (50, 150 i 200 minˉ¹) i početnih koncentracija (10, 20 i 30 g Lˉ¹) glukoze ili ksiloze u podlozi na prinos 2,3-butandiola te utjecaj acetata na proizvodnju 2,3-butandiola. Soj DSM 742 ne raste u rasponu pH vrijednosti od 3 do 5 jedinica. Najveći prinos 2,3-butandiola od 6,44 g Lˉ¹ i produktivnost 0,215 g Lˉ¹ hˉ¹ postignuti su tijekom uzgoja provedenog pri 50 minˉ¹, početnoj koncentraciji glukoze od 20 g Lˉ¹ i pH vrijednosti od 6,5 jedinica. Uzgojem bakterije P. polymyxa DSM 742 na 20 g Lˉ¹ pri 200 minˉ¹ do eksponencijalne faze te smanjenjem na 50 minˉ¹ nakon dodatka 5 g Lˉ¹ acetata postignut je prinos 2,3-butandiola od 8,72 g Lˉ¹ uz produktivnost 0,242 g Lˉ¹ hˉ¹.The increase in demand for 2,3-butanediol has prompted research of biotechnological production of 2,3-butanediol with the aim of replacing existing chemical production. In this work, the production of 2,3-butanediol with the bacterium Paenibacillus polymyxa DSM 742 was investigated, observing growth at different pH values (3, 4, 5 and 6,5), the influence of mixing (50, 150, 200 minˉ¹), initial concentrations (10, 20, 30 g Lˉ¹) of glucose or xylose in the broth and effect of acetate on the yield of 2,3-butanediol. It was observed that strain DSM 742 does not grow between 3 and 5 pH units. Highest yield of 2,3-butanediol (6,44 g Lˉ¹) and productivity (0,215 g Lˉ¹ hˉ¹) were achieved at 50 minˉ¹, initial concentration of glucose 20 g Lˉ¹ and 6,5 pH units. By culturing P. polymyxa DSM 742 at 20 g Lˉ¹and 200 minˉ¹ until the exponential phase and reducing to 50 minˉ¹ after the addition of 5 g Lˉ¹ acetate, a yield of 8,72 g Lˉ¹ was achieved with productivity 0,242 g Lˉ¹ hˉ¹
