Angiotensin Converting Enzyme and Angiotensin II Type 1 Receptor Gene Polymorphisms in Association with Preeclampsia in Turkish Women

Yüksek Lisans TeziPreeklampsi idrarda önemli miktarda proteinin bulunması ile ilişkili olarak hamilelikte hipertansiyonun ortaya çıktığı tıbbi bir durumdur. Hassas kadınların maternal kan damarlarında endotel disfonksiyona neden olan plasentadan gelen maddelerin olduğu muhtemel görülmektedir. Renin anjiyotensin sisteminin preeklampsinin patofizyolojisinde bir rol oynadığı belirtilmiştir. Renin anjiyotensin sistemi angiotensinojen, renin, anjiyotensin I, anjiyotensin I-dönüştürücü enzim, anjiyotensin II ve anjiyotensin II reseptör tip 1,2,3 ve 4 genlerinden oluşur. Anjiyotensin dönüştürücü enzim anjiyotensin I'in karboksi-terminal dipeptidine bağlanır, fizyolojik olarak aktif oktapeptid anjiyotensin II'yi açığa çıkarır. Anjiyotensin II vasküler tonu ayarlamada kilit bir rol oynayan vazokonstriktif etkili bir moleküldür. Bu çalışmanın amacı Türk kadınlarındaki preeklampsi hastalarında anjiyotensin dönüştürücü enzim ve anjiyotensin II tip 1 reseptör gen polimorfizmlerinin sıklığını araştırmaktır. Çalışma 75 preeklampsili hasta ve 75 kontrol grubu içermektedir. Anjiyotensin dönüştürücü enzim gen polimorfizmi polimeraz zincir reaksiyonu kullanılarak araştırılmış, anjiyotensin II tip 1 reseptör gen polimorfizmleri de polimeraz zincir reaksiyonu ve restriksiyon fragman uzunluk polimorfizmi yöntemleri kullanılarak belirlenmiştir. Çalışmamızda anjiyotensin dönüştürücü enzim ve anjiyotensin II tip 1 reseptör gen polimorfizmlerinin Türk kadınlarında preeklampsi gelişmesinde genetik risk faktörleri olmadıkları belirlendi.AbstractPreeclampsia is a medical condition in which hypertension arises in pregnancy in association with significant amounts of protein in the urine. It appears likely that there are substances from the placenta that can cause endothelial dysfunction in the maternal blood vessels of susceptible women. The renin angiotensin system has been implicated to play a role in the pathophysiology of preeclampsia. This system comprises the angiotensinogen, renin, angiotensin I, angiotensin I-converting enzyme, angiotensin II and angiotensin II receptor types 1,2,3 and 4 genes. Angiotensin converting enzyme cleaves the carboxy-terminal dipeptide of angiotensin I, releasing the physiologically active octapeptide angiotensin II. Angiotensin II is a potent vasoconstrictive molecule that plays a key role in modulating vascular tone. The aim of this study was to investigate the frequency of angiotensin converting enzyme and angiotensin II type 1 receptor gene polymorphisms in preeclampsia patients in Turkish women. The study involved 75 patients with preeclampsia and 75 control subjects. The angiotensin converting enzyme gene polymorphism was investigated using polymerase chain reaction, and the angiotensin II type 1 receptor gene polymorphism was identified using polymerase chain reaction and restriction fragment length polymorphism assay. Our results show that angiotensin converting enzyme and angiotensin II type 1 receptor gene polymorphisms were not genetic risk factors for preeclampsia in Turkish women
