Influence of alpha cypermethrin and difenoconazole pesticides on European green toad tadpoles (Bufotes viridis L.)

Na globalnoj razini, jedna od najugroženijih skupina životinja su vodozemci, a jedan od uzroka je i prekomjerno korištenje pesticida. Upotreba pesticida, uključujući alfa cipermetrin i difenkonazol, na oranicama u blizini vodenih površina predstavlja veliku opasnost za punoglavce zbog njihovog ispiranja oborinama ili tijekom poplava. Stoga, ciljevi ovog istraživanja bili su utvrditi kakav utjecaj imaju pesticidi alfa cipermetrin i difenkonazol na oksidativni stres punoglavaca zelene krastače (Bufotes viridis L.) analizom tri molekularna biomarkera stresa (aktivnost katalaze, sadržaj proteinskih karbonila te aktivnost transportnog sustava elektrona (ETS) na tri različite temperature (20°C, 24°C i 28°C)), utvrditi utjecaj pesticida na mortalitet i masu punoglavaca te koji je pesticid štetniji. Rezultati su pokazali povećanu aktivnost katalaze i karbonilnih skupina pri nekoliko koncentracija pesticida u odnosu na kontrolu. Mjerenja aktivnosti ETS-a pokazala su da nema razlike u aktivnosti na različitim temperaturama, ali da je aktivnost ETS-a veća kod difenkonazola. Mortalitet je utvrđen samo kod punoglavaca tretiranih difenkonazolom, dok je smanjenje mase utvrđeno kod oba pesticida. Iz rezultata možemo zaključiti da ovi pesticidi imaju negativne učinke na punoglavce povećavajući oksidativni stres i smrtnost te smanjujući masu punoglavca. Utvrđeno je da je difenkonazol štetniji pesticid zbog povećane smrtnosti, veće aktivnosti ETS-a te povećane prisutnosti karbonilnih skupina: Declinesin amphibian populations over the past few decades have become more significant and accelerating. One of the reasons for their decline is the excessive use of pesticides. Therefore, the aims of this study were to determine the effects of alpha cypermethrin and difenoconazole on the oxidative stress in European green toad tadpoles (Bufotes viridis L.) by analyzing three molecular biomarkers of stress (catalase activity, protein carbonyl content and electron transport system activity (ETS) at three different temperatures (20°C, 24°C, 28°C)), determining the effect of these pesticides on mortality and tadpoles mass and to determine which pesticide has more harmful effect on tadpoles. Results showed increased catalase activity and carbonly content in several concentrations compared to the control group. Measurements of ETS activity showed no difference in activity at different temperatures, but ETS activity is higher with difenconazole treatment. Mortality was observed only in tadpoles treated with difenoconazole, while mass decrease was observed in tadpoles treated with both pesticides. Based on our results we can conclude that these pesticides have negative effects on tadpoles and that difenoconazole is a more harmful pesticide due to increased mortality, higher ETS activities and increased presence of carbonyl groups
