Investigation of the preconcentration conditions of Cd(II), Cu(II) and Pb(II) ions with solid phase extraction by using fungus (Mucor pusillus)

Bu çalışmada, Mucor pusillus immobilize edilmiş Amberlit XAD-4 üzerinde Cd(II), Cu(II) ve Pb(II) iyonlarının katı faz özütleme yöntemi ile zenginleştirilme şartları araştırıldı. Metal iyonların tayinleri yüksek çözünürlüklü alevli atomik absorpsiyon spektroskopi cihazı ile yapıldı. Çalışılan metal iyonların geri kazanma verimine örnek çözeltisinin ortamının pH'sının, geri alma çözeltilerinin türü ve derişimlerinin, çözelti akış hızının ve çözelti hacminin etkisi incelendi. Belirlenen en uygun deneysel şartlarda Cd(II), Cu(II) ve Pb(II) iyonlarının geri kazanma verimi %95 güven seviyesinde sırasıyla, %99±3, %97±2 ve %96±2 olarak, bulundu. Yöntemin kesinliği, çalışılan iyonlar için gözlenebilme sınırı, bilinen miktarda analat eklenerek ve standart referans maddeler kullanılarak yöntemin doğruluğu belirlendi. Geliştirilen yöntem çeşme suyu, Kızılırmak suyu ve domates yaprağı örneklerinde, çalışılan metal iyonların tayinine uygulandı. Gerçek örnekler için bağıl standart sapma ve bağıl hata değerleri en fazla %8 olarak bulundu.In this study, preconcentration conditions of Cd(II), Cu(II) and Pb(II) ions for Mucor pusillus immobilized on Amberlite XAD-4 by solid phase extraction method were investigated. Mucor pusillus was studied as microorganism. The determination of the elements was carried out by flame atomic absorption spectrometry. The effect of pH, flow rate and volume of sample solution on the recovery of the studied ions was investigated. The recoveries Cd(II), Cu(II) and Pb(II) ions were found as 99± 3%, 97±2 % and 96±2 % respectively for Mucor pusilus immobilized on Amberlite XAD-4, under the optimum conditions determined. The accuracy of the method by using addition samples, precision of the method and limit of detection for the studied ions were determined. The proposed method was applied to tap water, Kızılırmak river water and tomato leaves for the determination of the analytes. Relative standard deviation and relative error were found as about 8 %, respectively for real samples
