Biological calcification: formation and crystal growth of calcium carbonate in microalgae

In numerous applications in both medicine and industry, the formation of deposits on process equipment and parts consisting of both organic and (mainly) inorganic salts, including calcium carbonate, is the cause of operational problems. Mineral deposits associated with the development of microorganisms (biological deposits) are complicated forms which need special attention. The formation calcium carbonate, associated with the growth of biological substrates, addressed to in the present work, is a challenging research topic. In the first part of this work, the mechanism of spontaneous precipitation of calcium carbonate in supersaturated solutions and the effect of the presence of heavy metals was investigated. The free drift spontaneous precipitation and the corresponding repetitive study at constant supersaturation, were used to establish the stability diagram of the calcium carbonate supersaturated solutions.Precipitation in supersaturated solutions, corresponding to the unstable solutions domain, yielded measurements of the induction time, lapsed from the preparation of the supersaturated solutions to the onset of the precipitation of the calcium carbonate, and of the rate of precipitation. From the dependence of the rate of precipitation on the corresponding saturation ratio with respect to calcite (5.89<SRvaterite<9.12, 0.10 M NaCl), it was possible to identify that the rate determining step was surface diffusion of the growth units to the growing calcium carbonate supercritical nuclei. The rate–supersaturation ratio data, for supersaturated solutions with ionic strength 0.1 and 0.15M, gave satisfactory fit to the polynuclear crystal growth model. Fit of the data to the Avrami model gave apparent order value for the precipitation, n, gave different values, for low (0.47<SRvaterite<0.58) and for higher relative supersaturation values (1.42<SRvaterite<2.01). However, the Avrami model, is concerned only with the growth in size, of a fixed number of nuclei, as a function of time. The surface energy, calculated from the plots according to this model, was 114mJ-1∙m-and the number of modules, which constitute the critical nuclei, equal to 4. The influence of the presence of Acutodesmus Obliquus microalgae growth medium in the supersaturated solutions, on the spontaneous precipitation of calcium carbonate (0.15M NaCl, 25oC, pH=8.50) was studied next using stable supersaturated solutions according to the stability diagram. Supersaturated solutions (11.48<SRcalcite<53.70), in which the culture medium was used as the matrix, suppressed completely the precipitation of CaCO3 and the solutions remained stable. However, modification of the supersaturated solutions to contain 1% w/w of medium (11.48<SRcalcite<79.43), resulted in an acceleration of the precipitation, as much as by 85% of the rate value in the absence of microalgae nutrients medium. The presence of the nutrient medium in the supersaturated solutions resulted in the stabilization of vaterite.The effect of the presence of heavy metals (Zn, Ni, Cd, Cu) in the supersaturated solutions during spontaneous precipitation of CaCO3 was studied next. In the presence of metals, at a concentration of 20µM in the supersaturation ratio range 26.91<SRcalcite<79.43, the rate of precipitation of calcium carbonate was drastically reduced (inhibition of 60% in the presence of Ni and Zn), and the corresponding induction times were significantly longer. In the presence of metals in the supersaturated solutions, higher surface energy values for the precipitated solid, were calculated. The presence of the metals investigated at the corresponding concentration range, did not have any effect on the precipitation mechanism. The presence of zinc in the supersaturated solutions (ionic strength 0.1M NaCl & 47.86<SRcalcite<112.2), shifted viiitheir stability domain to higher relative supersaturation values. The rate of calcium carbonate precipitation in the presence of zinc was reduced by 98%, with respect to the corresponding in its absence. In the presence of zinc, the aragonite was stabilized. The second part of the present work was concerned with the study of heterogeneous precipitation of CaCO3, following the inoculation of the supersaturated solutions with calcite and biological substrates. In stable supersaturated solutions, (SRcalcite=1.66-9.55), for ionic strength 0.15 M NaCl, the rate of crystal growth increased with supersaturation following a quadratic mode. Keeping the supersaturation value constant in the solutions, the precipitation rate was proportional to the total surface of the substrate, confirming that the number of active crystallization centers is constant. The presence of zinc on the supersaturated solutions at concentrations up to 3∙10-5M for ionic strength 0.1M (SRcalcite=7.41) and 0.15M NaCl (SRcalcite=3.55) reduced the rate of crystal growth of calcium carbonate. The rate reduction was higher with, increasing the zinc concentration, reaching 90% of the corresponding rate in its absence. The crystal growth rates of calcium carbonate, measured as a function of zinc concentration, were fitted to a model based on the assumption that the rate reduction is due to adsorption and poisoning of active sites for crystal growth. The adsorption was assumed to take place according to the Langmuir model. The presence of zinc in the supersaturated solutions resulted in a complete inhibition of the crystallisation of calcium carbonate, even at concentrations below the concentration, needed for complete monolayer coverage of the substrate from the zinc chemicals species. The identification of the crystalline solid formed on the substrate confirmed the absence of metal oxide and/or hydroxide. The study of zinc absorption in calcite showed that zinc’sabsorption in the solid substrate is initially (the first 10-20minutes) rapid and slower thereafter, approaching equilibrium, corresponding to the maximum amount of zinc that may be absorbed. The concentration measurements of zinc in solutions as a function of time, during adsorption, gave a good fit to the pseudo-first order model. Equilibrium adsorption data gave satisfactory fir to the Langmuir isotherm presence inthe supersaturated solutions, resulted in a reduction in the rate of precipitation of calcium carbonate (by 60%). Measurements of nickel adsorption on calcite confirmed the high affinity of the calcium substrate for the metal. Finally, the absorption of cadmium on calcite was slower, in comparison with zinc and nickel. It has also been shown that calcite exhibited selectivity for the elimination of cadmium from solutions reaching 85% of the initial concentration. The effect of biological substrates, on the nucleation and growth of calcium carbonate, was studied by the inoculation of stable supersaturated solutions with substrates prepared from the initial culture of the microalgae Acutodesmus obliquus. The microalgae was cultured in Basal Bold’s Medium, in order to measure its rate of growth by measurements of the concentration of chlorophyll produced and the cell density as a function of time. The identification of the chemical composition of the microalgae confirmed the presence of carboxyl and amino groups. The formation of calcium carbonate from supersaturated solutions (7.94<SRcalcite<104.71) in culture has been shown to depend on cell density. Higher precipitation rates of calcium carbonate were measured by increasing the cell density of cultures ofmicroalgae. The precipitation rate of CaCO3 depends on the illumination of the solutions and the presence of medium for microalgae growth. These parameters resulted in an acceleration of precipitation rate of calcium carbonate. The measurement of carbonate concentration ixconfirmed the growth of microalgae by photosynthesis during formation of calcium carbonate. The importance of photosynthesis in the precipitation of calcium carbonate was also confirmed by the study of substrates of microalgae, which were dried in the air at 25°C. The presence of these substrates in supersaturated solution with respect to calcium carbonate, has resulted in the increase of the precipitation rate, in comparison with the respective rates in the presence of active microalgae culture. Inoculation of stable supersaturated solutions with microalgae cultures air dried at 70°C, did not induce precipitation. Composite substrates of microalgae withcalcite, precipitated in air-dried microalgae, were more effective in inducing doubling of the rate of precipitation, in comparison with the corresponding rate obtained with calcite. In the last part of the study on the effect of biological substrates on the crystal growth of calcium carbonate, the effect of heavy metals was investigated. The active microalgae culturein the presence of calcite, increased zinc absorption. Similar results were obtained for calciteprecipitated on air-dried microalgae. The precipitation of calcium carbonate in supersaturated solutions in the presence of zinc and active microalgae was reduced by 80% (of the respective rate in their absence), while the growth rate of microalgae culture was reduced. Nickel was not absorbed into substrates of air-dried cultures of microalgae. However, in solutions, in the presence of nickel and active microalgae culture, the rate of precipitation of calcium carbonate was reduced by 88%. The presence of cadmium in the supersaturated solutions with respect to calcium carbonate, upon inoculation with air dried microalgae culture, cancelled CaCO3precipitation. The presence of Ni, Zn, Cd and Cu (CMe=10-5M), in the supersaturated solutions resulted to significant reduction of their concentration in the respective supersaturated solutions. The effect of the presence of cadmium in the supersaturated solutions was significant. The binding of metals on the test substrates may be attributed to surface complexation. Calculations of the capacitance of the substrate electrolyte double layer in the presence of metals from potentiometric titrations data on dry microalgae substratescorroborated this suggestion.Σε πολλές εφαρμογές τόσο στην ιατρική όσο και στην βιομηχανία ο σχηματισμός εναποθέσεων οι οποίες αποτελούνται τόσο από οργανικά αλλά και (κυρίως) από ανόργανα άλατα, όπως το ανθρακικό ασβέστιο, στον αντίστοιχο εξοπλισμό, αποτελεί πηγή προβλημάτων στην καλή του λειτουργία. Δύο είναι οι κύριες κατηγορίες εναποθέσεων: οι οφειλόμενες στην ανάπτυξη μικροοργανισμών (βιολογικής προέλευσης εναποθέσεις) και οι εναποθέσεις ανόργανων σωματιδίων. Ο τρόπος με τον οποίο επιδρούν τα βιολογικά υποστρώματα στην καταβύθιση ανόργανων σωματιδίων, όπως το ανθρακικό ασβέστιο, αποτελεί θέμα προς μελέτη το οποίο παρουσιάζει σημαντικές προκλήσεις. Στο πρώτο μέρος της παρούσας εργασίας διερευνάται ο μηχανισμός της αυθόρμητης καταβύθισης του ανθρακικού ασβεστίου σε υπέρκορα διαλύματά του, καθώς και η σημασία της παρουσίας σε αυτά βαρέων μετάλλων. Μέσω πειραμάτων τόσο μη ελεγχόμενων συνθηκών (free drift) αλλά και σταθερής σύστασης, κατασκευάσθηκε το διάγραμμα σταθερότητας των υπέρκορων ως προς το ανθρακικό ασβέστιο διαλυμάτων. Από την περιοχή τιμών υπερκορεσμού, οι οποίες καθορίζουν το χρόνο παραμονής των διαλυμάτων σε σταθερή κατάσταση και από τους χρόνους επαγωγής, οι οποίοι μεσολαβούσαν από την παρασκευή των υπέρκορων διαλυμάτων μέχρι την έναρξη της καταβύθισης του ανθρακικού ασβεστίου, επιλέχθηκαν οι συνθήκες για τη μελέτη του μηχανισμού της καταβύθισης του ανθρακικού ασβεστίου. Όπως διαπιστώθηκε, αυξανομένου του υπερκορεσμού, ο ρυθμός καταβύθισης αυξήθηκε ενώ μειώθηκε ο αντίστοιχος χρόνος επαγωγής. Σε ιοντική ισχύ 0.1Μ με NaCl, επικράτησε ο σχηματισμός βατερίτη, σε αντίθεση με την καταβύθιση σε υπέρκορα διαλύματα στα οποία η ιοντική ισχύς ήταν 0.15Μ, στα οποία σχηματίστηκε αποκλειστικά ασβεστίτης. Σε υπέρκορα διαλύματα στην περιοχή τιμών υπερκορεσμού 5.89<SRvaterite<9.12 και ιοντική ισχύ 0.1Μ NaCl από την εξάρτηση του ρυθμού καταβύθισης, από τον σχετικό υπερκορεσμό των διαλυμάτων, και από την προσαρμογή των αντίστοιχων δεδομένων σε μοντέλα κρυσταλλικής ανάπτυξης, εξήχθη το συμπέρασμα, ότι η κινητική καταβύθισης του ανθρακικού ασβεστίου καθορίζεται από την επιφανειακή διάχυση. Η προσαρμογή των δεδομένων των μετρήσεων του ρυθμού καταβύθισης από την τιμή του σχετικού υπερκορεσμού, ήταν πολύ καλή στο πολυπυρηνικό πρότυπο κρυσταλλικής ανάπτυξης. Στη σειρά αυτή των μετρήσεων, έγινε προσαρμογή των τιμών προστιθέμενου όγκου για διατήρηση σταθερού υπερκορεσμού, στο μοντέλο Avrami, για τον υπολογισμό εκ νέου του ρυθμού αυθόρμητης καταβύθισης του CaCO3. Όπως φάνηκε οι ‘νέοι’ ρυθμοί, οι οποίοι υπολογίστηκαν ήταν αρκετά μικρότεροι σε σχέση με τους αντίστοιχους, οι οποίοι υπολογίστηκαν απευθείας από τα διαγράμματα προστιθέμενου όγκου συναρτήσει του χρόνου. Η τιμή της φαινόμενης τάξης του ρυθμού, n, διέφερε κατά δύο μονάδες, από την αρχικά υπολογισμένη, τόσο για χαμηλές (0.47<σvaterite<0.58) όσο και για υψηλότερες τιμές σχετικού υπερκορεσμού (1.42<σ vaterite<2.01). Η απόκλιση αυτή πιθανόν να οφείλεται στις βασικές υποθέσεις του μοντέλου Avrami, με τη βοήθεια του οποίου περιγράφεται η ανάπτυξη ενός σταθερού πυρήνα συναρτήσει του χρόνου. Οι μετρήσεις του ρυθμού καταβύθισης στην περιοχή τιμών υπερκορεσμού 11.48<SRcalcite <79.43, σε ιοντική ισχύ 0.15Μ NaCl (Vreactor=500mL), έδειξαν ο μηχανισμός καταβύθισης του CaCO3 περιγράφεται επίσης από το πολυπυρηνικό πρότυπο. Η επιφανειακή ενέργεια, σύμφωνα με το συγκεκριμένο πρότυπο υπολογίστηκε 114mJ∙m-2 και ο αριθμός των δομικών μονάδων, οι οποίες συγκροτούν τον κρίσιμο πυρήνα, ίσος με 4. Στη συνέχεια μελετήθηκε η επίδραση της παρουσίας του θρεπτικού μέσου ανάπτυξης της μικροάλγης στα υπέρκορα διαλύματα, στην αυθόρμητη καταβύθιση του iv ανθρακικού ασβεστίου (0.15Μ NaCl, 25οC, pH=8.50). Η αντικατάσταση του διαλύτη (νερό) με θρεπτικό μέσο, για τιμές υπερκορεσμού 11.48<SRcalcite<53.70, έδειξαν να αναστέλλουν την αυθόρμητη καταβύθιση του CaCO3, καθώς τα υπέρκορα διαλύματα παρέμειναν σταθερά. Ωστόσο, τροποποίηση των υπέρκορων διαλυμάτων ώστε να περιέχουν 1% w/w θρεπτικού μέσου (11.48<SRcalcite<79.43), είχε ως αποτέλεσμα την επιτάχυνση της καταβύθισης ακόμη και κατά 85% της τιμής του ρυθμού σε σύγκριση με την αντίστοιχη απουσία του θρεπτικού μέσου. Η παρουσία του θρεπτικού μέσου, είχε ως αποτέλεσμα τη σταθεροποίηση του βατερίτη. Ακολούθως μελετήθηκε η επίδραση της παρουσίας βαρέων μετάλλων (Zn, Ni, Cd,Cu) στα υπέρκορα διαλύματα κατά την αυθόρμητη καταβύθιση του CaCO3. Παρουσία μετάλλων, σε συγκέντρωση 20μΜ στην περιοχή τιμών υπερκορεσμού 26.91<SRcalcite<79.43, διαπιστώθηκε σημαντική μείωση του ρυθμού καταβύθισης του ανθρακικού ασβεστίου (αναστολή 60% παρουσία Ni και Ζn) και αύξηση του χρόνου επαγωγής. Η μεγαλύτερη τιμή επιφανειακής ενέργειας του καταβυθιζόμενου στερεού, που υπολογίστηκε, επιβεβαιώνει τον λόγο επιβράδυνσης της πυρηνογένεσης. Ωστόσο, η μείωση αυτή στον ρυθμό δεν είχε καμία επίδραση στον μηχανισμό καταβύθισης, ενώ στο στερεό που συλλέχθηκε δεν ταυτοποιήθηκε καμία φάση οξειδίων ή/και υδροξειδίων του μετάλλου. Παρουσία ψευδαργύρου κατά την εκτενέστερη μελέτη της επίδρασης της παρουσίας του στα υπέρκορα διαλύματα (ιοντική ισχύς 0.1Μ NaCl & 47.86<SRcalcite<112.2) διαπιστώθηκε η μετατόπιση της περιοχής σταθερότητας σε μεγαλύτερες τιμές σχετικού υπερκορεσμού. Ο ρυθμός καταβύθισης του ανθρακικού ασβεστίου παρουσία ψευδαργύρου μειώθηκε κατά 98%, ως προς τον αντίστοιχο απουσία του. Παρουσία ψευδαργύρου στα υπέρκορα διαλύματα, σταθεροποιήθηκε ο αραγωνίτης, πιθανώς λόγω της επιβράδυνσης της μετατροπής του σε ασβεστίτη. Το δεύτερο μέρος της εργασίας αφορούσε τη μελέτη της ετερογενούς καταβύθισης του CaCO3, απουσία και παρουσία βιολογικών υποστρωμάτων. Σε σταθερά υπέρκορα διαλύματα, τα οποία αντιστοιχούσαν στην περιοχή τιμών υπερκορεσμού SRcalcite=1.66-9.55, για ιοντική ισχύ 0.15Μ NaCl σε συνθήκες σταθερής σύστασης, ο ρυθμός κρυσταλλικής ανάπτυξης αυξήθηκε παραβολικά αυξανομένου του υπερκορεσμού. Διατηρώντας σταθερή την τιμή του υπερκορεσμού στα υπέρκορα διαλύματα, ο ρυθμός καταβύθισης ήταν ανάλογος της ολικής επιφανείας του υποστρώματος, γεγονός το οποίο σημαίνει, ότι ο αριθμός των ενεργών κέντρων κρυστάλλωσης είναι σταθερός. Στην συνέχεια, μελετήθηκε η επίδραση της παρουσίας ψευδαργύρου στα υπέρκορα διαλύματα σε συγκεντρώσεις έως 3∙10-5 Μ, για ιοντική ισχύ 0.1Μ (SRcalcite=7.41) και 0.15Μ NaCl (SRcalcite=3.55). Οι μετρήσεις έδειξαν ότι ο ρυθμός κρυσταλλικής ανάπτυξης μειώθηκε αυξανομένης της συγκέντρωσης ψευδαργύρου, με το μέγιστο ποσοστό αναστολής της καταβύθισης παρουσία ψευδαργύρου, να φτάνει το 90%. Οι ρυθμοί κρυσταλλικής ανάπτυξης του ανθρακικού ασβεστίου, οι οποίοι μετρήθηκαν, συναρτήσει της συγκέντρωσης του ψευδαργύρου, προσαρμόσθηκαν σε μοντέλο το οποίο βασίζεται στην υπόθεση ότι η μείωση του ρυθμού οφείλεται στην προσρόφηση και στη δηλητηρίαση των ενεργών κέντρων κρυστάλλωσης. Η προσρόφηση αυτή περιγράφεται από την ισόθερμο Langmuir. Η παρουσία ψευδαργύρου στα υπέρκορα διαλύματα, είχε ως αποτέλεσμα την πλήρη αναστολή της κρυστάλλωσης του ανθρακικού ασβεστίου, ακόμα και σε συγκεντρώσεις μικρότερες από την συγκέντρωση, η οποία απαιτείται για πλήρη μονοστρωματική κάλυψη του υποστρώματος από τα χημικά είδη του ψευδαργύρου, τα οποία ροφούνται. Η ταυτοποίηση του κρυσταλλικού στερεού, το οποίο σχηματίσθηκε στο υπόστρωμα, έδειξε ότι δεν σχηματίζεται κάποιο οξείδιο ή/και υδροξείδιο του μετάλλου. Η μελέτη της ρόφησης του vψευδαργύρου σε ασβεστίτη, έδειξε, ότι η ρόφηση του Zn στο στερεό υπόστρωμα γίνεται αρχικά (τα πρώτα 10-20 λεπτά) με ταχύτατο ρυθμό και βραδύτερα στη συνέχεια, προσεγγίζοντας την ισορροπία, η οποία αντιστοιχεί στη μέγιστη ποσότητα ψευδαργύρου, που είναι δυνατό να ροφηθεί. Η προσαρμογή των μετρήσεων της απομείωσης της συγκέντρωσης του ψευδαργύρου στα διαλύματα συναρτήσει του χρόνου, λόγω προσρόφησής του, σε κινητικά μοντέλα ρόφησης ήταν αρκετά καλή και η ισόθερμος Langmuir έδειξε να περιγράφει ιδανικά τον μηχανισμό ρόφησης στην κατάσταση ισορροπίας. Ακολούθησε η μελέτη της επίδρασης του νικελίου στην κρυσταλλική ανάπτυξη του CaCO3, σε συγκεντρώσεις παραπλήσιες προς τις αντίστοιχες του ψευδαργύρου. Η παρουσία του στα υπέρκορα διαλύματα, είχε ως αποτέλεσμα την μείωση του ρυθμού καταβύθισης του ανθρακικού ασβεστίου (κατά 60%). Μετρήσεις της ρόφησης Ni σε ασβεστίτη, επιβεβαίωσαν την συνάφεια του υποστρώματος ασβεστίτη προς το μέταλλο. Η προσαρμογή των μετρήσεων προσρόφησης στην κατάσταση ισορροπίας στην ισόθερμο Langmuir ήταν ικανοποιητική. Τέλος, η ρόφηση του το καδμίου ήταν βραδύτερη στην επιφάνεια του ασβεστίτη σε σύγκριση με τους αντίστοιχους ρυθμούς για τον ψευδάργυρο και το νικέλιο. Επίσης, αποδείχθηκε, ότι ο ασβεστίτης παρουσιάζει μεγαλύτερη επιλεκτικότητα ως προς την απομάκρυνση του καδμίου από διαλύματά του σε ποσοστό, το οποίο φθάνει ακόμα και το 85% της αρχικής συγκέντρωσης στα διαλύματα. Η μελέτη της επίδρασης των βιολογικών υποστρωμάτων που ακολούθησε βασίστηκε σε υποστρώματα, τα οποία παρασκευάστηκαν από την αρχική καλλιέργεια της μικροάλγης Acutodesmus obliquus. Η ανάπτυξη της στο θρεπτικό μέσο Basal Bold’s Medium, έγινε προκειμένου να μετρηθεί ο ρυθμός ανάπτυξής της, από μετρήσεις της συγκέντρωσης της παραγόμενης χλωροφύλλης και της κυτταρικής πυκνότητας συναρτήσει του χρόνου.
