Investigation of the effect of Toxoplasma gondii on ram fertilizing capacity

The aim of this study was to investigate the effect of experimental Toxoplasma gondii infection on ram sperm quality. Thirty-two five months old, pre-pubertal, rams were divided into 4 groups (n=8 per group). Group A was the control group; the remaining three groups (B, C, D) received per os 5000 mature oocysts of T. gondii. Group C received sulphadimidine (33 mg/kg; i.m. every 48h for 8 days) two months post-infection (p.i.) and Group D received the same drug twice (24h p.i. and 2 months later). Group B did not receive treatment. Blood samples were collected every 15 days to detect serum immunoglobulin IgG (ELISA). The animals of all groups were euthanized 4 months p.i. and epididymal sperm samples were analyzed for a) presence of parasite’s DNA (PCR), b) concentration (Neubauer haemocytometer chamber), viability and morphology (Eosin–Nigrosin staining), kinetics (computer–assisted sperm analysis system — CASA), membranes’ functional integrity (HOS Test) and DNA integrity (Acridine Orange Test). Furthermore, histopathological examination was performed on the testes. The IgG titres in infected animals raised 2 weeks p.i. and remained high for 4 months. Treatment with sulphadimidine 24h after experimental infection of pre-pubertal rams with T. gondii delayed the immune response for 1 week and additionally decreased the IgG values. Parasite’s DNA was detected in three sperm samples of the infected rams (12.5%) 15 weeks p.i. Significantly higher values were noticed in viability and spermatozoa membrane functional integrity in the control group compared to infected groups, level of significance p<0.05. Abnormal sperm was higher in groups C and D vs. A and C vs. B (p<0.05). Experimental Toxoplasmosis had no significant burden on the main sperm motility characteristics of rams and did not affect the spermatozoa chromatin integrity (DNA). Histopathology revealed similar findings with little variation among all infected groups, characterized mostly by increased interstitial connective tissue, non-purulent inflammation, and presence of seminiferous tubules with spermatogenic cell depletion, which increased gradually from D to C and B group. Toxoplasma cysts were not found in any of the tissue samples from the infected animals. In summary, this thesis led to the conclusion that the experimental infection of rams during the pre–pubertal period of age with mature oocysts of T. gondii degrades sperm’s quality at puberty, consequently and the fertilizing ability, while the administration of sulphadimidine cannot prevent this effect.Σκοπός αυτής της μελέτης ήταν η διερεύνηση της επίδρασης της πειραματικής μόλυνσης με Toxoplasma gondii στην ποιότητα του σπέρματος του κριού. Τριάντα δύο άνηβοι κριοί ηλικίας 5 μηνών χωρίστηκαν σε 4 ομάδες (n=8). Η ομάδα Α αποτέλεσε την ομάδα ελέγχου, ενώ οι υπόλοιπες τρεις ομάδες (Β, Γ, Δ) μολύνθηκαν από το στόμα με 5000 ώριμες ωοκύστεις του πρωτόζωου T. gondii. Στην ομάδα Γ χορηγήθηκε σουλφαδιμιδίνη (33mg/kg ΣΒ, i.m., ανά διήμερο, για 8 ημέρες) 2 μήνες μετά τη μόλυνση και στην ομάδα Δ η ίδια αγωγή δύο φορές (24 ώρες μετά τη μόλυνση και 2 μήνες αργότερα). Η ομάδα Β δεν έλαβε φαρμακευτική αγωγή. Δείγματα αίματος συλλέγονταν ανά 15 ημέρες για την ανίχνευση ειδικών αντισωμάτων IgG (ELISA). Τα ζώα όλων των ομάδων θανατώθηκαν 4 μήνες μετά τη μόλυνση και από τις επιδιδυμίδες τους συλλέχθηκαν δείγματα σπέρματος τα οποία υποβλήθηκαν σε α) ανίχνευση του παρασίτου με PCR, β) εξετάσεις ποιοτικών χαρακτηριστικών: πυκνότητα (αιμοκυτταρόμετρο Neubauer), ζωτικότητα και μορφολογία (χρώση Εωσίνης–Νιγροσίνης), κινητικότητα (computer–assisted sperm analysis system — CASA), βιοχημική λειτουργικότητα των μεμβρανών των σπερματοζωαρίων (δοκιμή HOS) και ακεραιότητα της δομής της χρωματίνης (DNA) του πυρήνα των σπερματοζωαρίων (δοκιμή πορτοκαλόχρωμης ακριδίνης). Επιπλέον, λήφθηκαν ιστοτεμάχια από τους όρχεις για ιστοπαθολογική εξέταση. Στα μολυσμένα ζώα ο τίτλος αντισωμάτων αυξήθηκε τη 2η εβδομάδα μετά τη μόλυνση και παρέμεινε υψηλός για 4 μήνες. Η χορήγηση σουλφαδιμιδίνης i.m. 24 ώρες μετά τη πειραματική μόλυνση άνηβων αρσενικών αμνών με T. gondii, επέφερε καθυστέρηση στην ανοσιακή απόκριση κατά 1 εβδομάδα, αλλά και χαμηλότερες τιμές ειδικών αντισωμάτων IgG. Το DNA του παρασίτου ανιχνεύθηκε σε 3 δείγματα σπέρματος μολυσμένων κριών (12,5%), 15 εβδομάδες μετά τη μόλυνση. Σημαντικά υψηλότερες τιμές παρατηρήθηκαν στη ζωτικότητα και στη βιοχημική λειτουργικότητα των μεμβρανών των σπερματοζωαρίων στην ομάδα ελέγχου σε σύγκριση με τις μολυσμένες ομάδες, επίπεδο σημαντικότητας p <0,05.
