Studies on gas-phase fragmentation of homolicorin alkaloids from Hippeastrum species by sequential mass spectrometry with electrospray ionization

Neste trabalho, as fragmentações de sete alcaloides homolicorínicos (AHL) foram investigadas empregando espectrometria de massas sequencial com ionização por eletrospray (ESI-MS/MS). As vias de fragmentação dos compostos selecionados em suas formas protonadas e sob condições de dissociação induzida por colisão (CID) foram associadas às suas respectivas estruturas. Além das relações estrutura-fragmentação, dados de espectrometria de massas de múltiplos estágios (MSn) e experimentos com troca de deutério foram também utilizados para a elucidação das vias de fragmentação. Os resultados dos estudos sobre a fragmentação dos alcaloides homolicorínicos 1-7 protonados revelaram que a maioria dos íons formados na energia de 20 eV são resultantes de eliminações de CH3, NH3, H2O, CO, CH2O, CH3NH2 e CH3OH. Os processos de fragmentação comuns incluem as perdas de H2O e de CH3 e a ocorrência de duas reações de retro-Diels-Alder competitivas, a partir dos quais é possível identificar alguns íons fragmentos diagnósticos de grupos funcionais específicos. Fragmentos formados por perdas consecutivas de dois radicais CH3 foram úteis para distinguir os compostos 8,9-dimetoxilados 5-7 de 1-4, que apresentam grupamento metilenodioxi fundido a C8 e C9. Eliminações consecutivas de três moléculas de H2O foram indicativas da presença de um grupo hidroxila em C3, enquanto a presença de um grupo metoxilo na mesma posição pôde ser diagnosticada por meio da perda de metanol diretamente do íon precursor. Por fim, as eliminações de NH3 e de CH3NH2 permitiram identificar a natureza do substituinte ligado diretamente ao nitrogênio (H ou CH3). Os dados obtidos deste estudo podem ser utilizados de forma conjunta para a elaboração de estratégias para a identificação desses compostos em extratos brutos de Amaryllidaceae utilizando a técnica HPLC-MS/MS sem a necessidade de padrões.In this work, the fragmentation of seven homolicorine-type alkaloids (HLA) was investigated by electrospray ionization tandem mass spectrometry (ESI-MS/MS). The fragmentation pathways of the selected alkaloids in their protonated forms and under collision-induced dissociation (CID) were associated to their respective structures. Besides the structure-fragmentation relationships, multiple-stage mass spectrometry (MSn) data and experiments with deuterium exchange were also employed to elucidate the fragmentation pathways. The results revealed that most of the product ions of protonated HLA 1-7 formed at the collision energy of 20 eV were the result of CH3, NH3, H2O, CO, CH2O, CH3NH2, and CH3OH eliminations. The common fragmentation processes include H2O and CH3 eliminations from the precursor ion, as well as the occurrence of two competitive retro-Diels-Alder (RDA) reaction. These RDA reactions enable the identification of some product ions that are diagnostic for specific functional groups. Two consecutive losses of CH3 were useful to distinguish the 8,9-dimethoxy compounds from those that display a methylenedioxy ring fused to C8 and C9. Consecutive eliminations of three H2O molecules were diagnostic for the hydroxy group at C3, whereas the presence of a methoxy group at the same position could be identified on the basis of a methanol elimination directly from the precursor ion. Finally, eliminations of NH3 andCH3NH2 were useful to identify the nature of the substituent at the nitrogen (H or CH3). Data obtained in this study can be used along for the elaboration of HPLC-ESI-MS/MS-based strategies for the identification of HLA in Hippeastrum crude extracts
