The subject of this thesis is the construction of an experimental set-up, and in particular of a tunable continuous-wave laser at 205nm, for the measurement of the ground state Lamb shift in atomic hydrogen. Chapter 1 deals with the Lamb shift from a historical point of view, and with the interest of its measurement, for metrology and test of quantum electrodynamics. Chapter 2 is devoted to the theory of the hydrogen atom. The principle of the experiment is based on the comparison of two frequencies which are in a ratio four : those of the two-photon transitions of 2S-6S or 2S-6D and 1S-3S.
