During this thesis, we have studied several biological processes involving transfert RNA. First, the study of the role of the nucleocapsid protein (NC) in the formation of the HIV-1 reverse transcription initiation complex enabled us to identify a strong and specific binding site for the NC onto the D-arm of tRNALys3 — the tRNA used as a primer by the reverse transcriptase for the cDNA(-) synthesis. This strong binding allows the melting of tertiary interactions between the T and D-arms of tRNALys3 and could also be a key factor driving the tRNALys3/viral RNA hybrid formation.
