This work of thesis concerns the use of new lipidic nanocarriers (LNCs) for oral administration. This system should make it possible to encapsulate anticancer drug and to improve its bioavailability after oral administration. In the first part of our work, in vitro properties of paclitaxel-loaded LNCs were studied in view of oral administration. The gastrointestinal stability was demonstrated after contact with in vitro simulated media. Moreover, studies of transport through the intestinal epithelium (Caco-2 cell model) showed that LNCs improve paclitaxel transport by active endocytic processes, especially via caveolae- and clathrin-mediated endocytosis and located in the microenvironment of the P-gp.
