In the transformation to a cancerous phenotype, cells often develop resistance to chemotherapeutic drugs and ionizing radiation used for cancer therapy. The P13-K/Akt pathway is a pro-growth, anti-apoptotic pathway downstream of receptor tyrosine kinases that is frequently dysregulated in tumor cells. Our hypothesis was that inhibition of PI3-K/Akt by adenoviral-delivery of the tumor suppressor PTEN would radiosensitize human tumor cell lines in vitro. Three cell lines were radiosensitized by our Ad-PTEN treatment as determined by clonogenic survival.
