Influence of the temperature factor on the distribution of NO-positive neurons in the hypothalamus of birds and mammals

In the brain nitrogen oxide (NO) acts as a messenger molecule in differentiation, synaptic plasticity and neurotoxicity. NO is produced by activation of nitric oxide synthase (NOS). The marker for NOS-positive neurons is nicotinamide adenine dinucleotide phosphate-diaphorase (NADPH-d). The aim of the study was to investigate the influence of the temperature factor on the distribution of NADPH-d-positive neurons in the hypothalamus of birds and mammals. In the experimental part of work were used adult domestic fowls (Gallus gallus) and rats (Rattus norvegicus). The animals were kept in a specially fitted thermal control chamber at a temperature + 22,0 ± 0,5 °С (control). The overheating and cooling of the animals were carried out in the thermal chamber at a temperature + 37,0 ± 0,5 °С and + 12,0 ± 0,5 °С, correspondingly, during 10 hours. To study the influence of the temperature factor on the development of NO-positive neurons of the anterior hypothalamus in the prenatal ontogeny of homoiothermal organisms experiments were carried out in duck embryos on E20, E23, E28 and E33. Three experimental series were performed: incubation at a temperature + 37,0 ± 0,5 °С (control), incubation at a temperature + 39,0 ± 0,5 °С during 3 hours (warm stimulation) and incubation at a temperature + 34,0 ± 0,5 °С during 3 hours (cold stimulation) on the respective experimental day. In the study was used the method of identification of neurons containing NADPH-d developed by Scherer-Singler et al (Scherer-Singler U., Vincent S.R., Kimura H., McGeer E.G. // J. Neurosci. Methods. - 1983. - Vol.9, №3. - P.229-234) in the revision of Hope and Vincent (Hope B.T., Vincent S.R. // J. Histochem. Cytochem. - 1989. - Vol.37. - P.653-661). In the study of serial sections of the hypothalamus of fowls and rats subjected to overheating the density of NADPH-d-positive neurons was found to be increased compared with the control animals in the lateral preoptic area to 16,37 % and 25,66 % (p < 0,05), in the supraoptic nucleus to 20,32 % and 17,53 % (p < 0,05) and in the paraventricular nucleus to 21,05 % and 21,04 % (p < 0,05), correspondingly. In the study of serial sections of the hypothalamus of fowls and rats subjected to cooling was found no statistically significant difference in the change of density of NADPH-d positive neurons compared with the control animals. It is also found that in the control and warm stimulated group NADPH-d positive neurons of the hypothalamus are detected in duck embryos starting from E23 during embryogenesis. In the cold stimulated group NADPH-d positive neurons of the hypothalamus were found already in embryos on E20. Also in the other age groups investigated, the activation of hypothalamic neuronal NOS was significantly increased after cold stimulation.В головном мозге оксид азота (NO) действует как передаточная молекула в дифференцировании нейронов, синаптической пластичности и нейротоксичности. NO вырабатывается при активации синтазы оксида азота (NOS). Маркером для NOS-положительных нейронов является никотинамидадениндинуклеотидфосфат-диафораза (NADPH-d). Целью исследования было изучение влияния температурного фактора на распределение NADPH-d- позитивных нервных клеток в гипоталамусе у птиц и млекопитающих. В экспериментальной части работы использованы взрослые особи кур домашних (Gallus gallus) и крыс (Rattus norvegicus). Животные содержались в специально оборудованном термостатируемом помещении при температуре воздуха + 22,0 ± 0,5 °С (контроль). Перегревание и охлаждение животных осуществляли в термокамере при температуре воздуха + 37,0 ± 0,5 °С и + 12,0 ± 0,5 °С соответственно в течение 10 часов. С целью изучения влияния температурного фактора на развитие NO-позитивных нейронов переднего гипоталамуса в пренатальном онтогенезе гомойотермных организмов эксперименты выполнены на 20-, 23-, 28- и 33-дневных эмбрионах утки. Проводилось три серии опытов: инкубация при температуре + 37,0 ± 0,5 °С (контроль), инкубация при температуре + 39,0 ± 0,5 °С в течение 3 ч (тепловая стимуляция) и инкубация при температуре + 34,0 ± 0,5 °С в течение 3 ч (холодовая стимуляция) в соответствующий эксперименталь ный день. В работе использован метод идентификации NADPH-d-содержащих нейронов разработанный Scherer-Singler et al (Scherer-Singler U., Vincent S.R., Kimura H., McGeer E.G. // J. Neurosci. Methods. - 1983. -Vol.9, №3. - P.229-234) в модификации Hope и Vincent (Hope B.T., Vincent S.R. // J. Histochem. Cytochem. -1989. - Vol.37. - P.653-661). При исследовании серийных срезов гипоталамуса кур и крыс, которые подвергались перегреванию, обнаружено увеличение плотности NADPH-d-позитивных нейронов по сравнению с контрольными животными в латеральной преоптической области на 16,3 7 % и 25,66 % (p < 0,05), супраоптическом ядре на 20,32 % и 17,53 % (p < 0,05) и паравентрикулярном ядре на 21,05 % и 21,04 % (p < 0,05) соответственно. При исследовании серийных срезов гипоталамуса кур и крыс, которые подвергались охлаждению, не обнаружена статистически достоверная разница изменения плотности NADPH-d-позитивных нейронов по сравнению с контрольными животными. Также установлено, что у эмбрионов утки в контрольной и стимулируемой теплом группе NADPH-d-положительные нейроны гипоталамуса обнаруживаются, начиная с 23 дня эмбрионального развития. В группе с холодовой стимуляцией NADPH-d-положительные нейроны гипоталамуса были обнаружены уже у 20-дневных эмбрионов. Кроме того, в других исследуемых возрастных группах, активация NOS нейронов гипоталамуса значительно увеличилась после холодовой стимуляции
