Production, partial purification, caracterization and application of thermo-stable alpha-amylase produced by bacterium.

Resumo: Os produtos da hidrólise do amido são, certamente, os ingredientes mais versáteis utilizados nas indústrias de alimentos, reconhecidos tanto pelas suas características tecnológicas e nutricionais quanto pela variedade de produtos deles obtidos. Dentre estes produtos, encontram-se os xaropes de açúcares e seus derivados, empregados tanto em processos fermentativos, quanto na produção de bebidas, de produtos dietéticos e sorvetes, na produção de gomas, sendo ainda aplicados nas indústrias farmacêuticas e químicas. Tendo as mesmas origens, estes produtos são diferenciados em função do processamento aplicado durante a liquefação do amido, sendo sua qualidade dependente da fonte de amido, das condições de processamento e das enzimas aplicadas. Entretanto, as enzimas apresentam como limitação a temperatura ótima de operação, sendo desnaturadas em altas temperaturas sem que ocorra a hidrólise do amido, o que representa um decréscimo na rentabilidade e conseqüente desperdício de material. Por esta razão, intensificaram-se as pesquisas para produção de alfa-amilase termoestável, capaz de manter sua atividade hidrolítica em processos industriais que utilizem altas temperaturas. Diversas linhagens do gênero Bacillus têm sido utilizadas para a síntese de alfa-amilase com ótimas atividades a altas temperaturas, mas estas enzimas ainda requerem a presença de íons cálcio (Ca2+) adicionados ao meio, o que significa mais uma etapa no processamento e um entrave na etapa subseqüente de refinação dos xaropes obtidos. O presente trabalho isolou uma linhagem identificada como Bacillus subtilis produtora de alfa-amilase termoestável, extracelular e com pH e temperaturas máximos de operação de 6 e 80°C. A enzima foi parcialmente purificada em coluna com resina butil, apresentando peso molecular estimado em 21,7 KDa. Os produtos de hidrólise de quatro diferentes tipos de amido (batata, milho, mandioca e trigo) foram analisados por cromatografia líquida e em papel e revelaram tratar-se de uma enzima sacarificante por produzir, majoritariamente, glicose, maltose e maltotriose em 12 horas de processo.Abstract: The products of starch hydrolysis are certainly the most versatile ingredients in use in the food industry, being recognized for their technological and nutritional features and for the variety of products derived from a simple raw material such as starch. These products include the sugar syrup and their derivatives, used in fermentative processes, in the production of soft drinks, dietary products, ice cream and gums, in addition to their application in the pharmaceutical and chemistry industries. These products have the same origin, being differentiated as a function of the processing applied during the liquefaction of starch. The quality depends on the starch source, the conditions of processing and the enzyme employed. However, all enzymes usually applied are limited by the operational temperature, being denatured by high processing temperature without completely hydrolyzing the starch, which may represent a decrease in the yield and consequent loss of material. Therefore, the search for thermo-stable alpha-amylase has been intensified, in order to find an enzyme capable of maintaining its hydrolytic activity during process that use high temperature. Some bacteria of the genus Bacillus have been used to synthesize alpha-amylase with excellent activity at high temperatures, but these enzymes require the presence of calcium ions (Ca2+), which means an additional processing step and subsequent refinement step of the final products. This research have isolated a bacteria identified as Bacillus subtilis that produces a thermo-stable alpha-amylase. The enzyme, showing maximum values for activity at pH and temperature of 6 and 80°C, was partially purified in a butyl-resin column and showed a molecular weight of 21,7KDa by filtration in Sephadex G-200 gel. The hydrolysis products from four different types of starch (potato, maize, cassava and wheat) were analyzed by both liquid and paper chromatography, showing the enzyme to be a saccarifiying one, producing, mainly, glucose, maltose and maltotriose after 12 hours of process
