Evaluation of different diagnostic techniques for bovine tuberculosis in tuberculin reagent animals

Bovine tuberculosis is an important infectious disease caused by Mycobacterium bovis. It is considered both a public health problem and the economic losses it generates. In the absence of an effective vaccine for this disease, control and eradication programs are based on the diagnosis through the intradermal tests in the live animal and slaughter of the positive animals. This work aimed to compare diagnostic methods for bovine tuberculosis in animals reactive to tuberculinization. Macroscopic examination, histopathological characterization, microbiological culture, identification of acid-fast bacilli (AFB), molecular and serological identification were performed. A total of 28 cattle were slaughtered in cold slaughter houses under state inspection in the state of Rio Grande do Sul. Blood samples were collectedat the time before the application of the tuberculin during the intradermal test. On the ot her hand, tissue samples with or without macroscopic lesions suggestive of tuberculosis were collectedat the time of slaughter. In the macroscopic evaluation, 14 animals (50%) presented lesions suggestive of tuberculosis. In the histological evaluation 12 samples (42.86%) had a characteristic granulomatous lesion. In the microbiological culture, 15 samples (53.57%) were isolated in the Stornebrink culture medium, and nine samples (32.14%) in the 7H9 medium. The identification of AFB wason lypossible in seven samples (25%). Already the molecular identification was determined in six samples (21.42%) and the serological in 26 samples (92.85%). Detailed macroscopic examination associated with the use of the serological test and together with the histopathological examination contributes both to the diagnosis of tuberculosis and to the success of a control and eradication plan.Sem bolsaA tuberculose bovina é uma importante doença infecciosa causada pelo Mycobacterium bovis. Ela é considerada tanto um problema na saúde pública quanto nas perdas econômicas que ela gera. Na ausência de uma vacina eficaz para essa doença, programas de controle e erradicação são baseados no diagnóstico através dos testes intradérmicos no animal vivo e abate dos animais positivos. Este trabalho teve como objetivo a comparação de métodos de diagnóstico para a tuberculose bovina em animais reagentes à tuberculinização. Foramrealizadosexame macroscópico, caracterização histopatológica, cultivo microbiológico, identificação de bacilos álcool-ácidos resistentes (BAAR), identificação molecular e sorológica. Um total de 28 bovinos foram abatidos em matadouros- frigorífico sob serviço de inspeção estadual no estado do Rio Grande do Sul. As amostras de sangue foram colhidas no momento antes da aplicação da tuberculina, durante a realização do teste intradérmico. Já as amostras de tecidos com ou sem lesão macroscópica sugestiva de tuberculose foram coletadas no momento do abate. Na avaliação macroscópica, 14 animais (50%) apresentaram lesão sugestiva de tuberculose. Na avaliação histológica 12 amostras (42,86%) apresentaram lesão granulomatosa característica. No cultivo microbiológico, 15 amostras (53,57%) foram isoladas no meio de cultura de Stornebrink, e nove amostras (32,14%) no meio 7H9. A identificação de BAAR só foi possível em sete amostras (25%). Já a identificação molecular foi determinada em seis amostras (21,42%) e a sorológica em 26 amostras (92,85%). O exame macroscópico detalhado, associado a utilização do ensaio sorológico e juntamente com o exame histopatológico contribui tanto para o diagnóstico da tuberculose quanto para o sucesso de um plano de controle e erradicação
