We demonstrate using biochemical and structural analysis that the type I-D CRISPR-Cas Cascade complex contains Cas11d small subunits expressed from an alternative start site within the cas10d gene. This small subunit is required for I-D Cascade to bind specific DNA targets. We found the alternative expression of the Cas11 small subunit is conserved in types I-D, I-B, and I-C systems, which represent nearly a quarter of sequenced CRISPR-Cas systems.
