The mRNA-based vaccine manufacturing process begins with the generation of a DNA template either through linearization of a plasmid with a restriction enzyme or through PCR amplification. The template should contain an RNA polymerase promoter, such as T7 or SP6, and the corresponding sequence for the mRNA construct. In vitro transcription ensues and the template is subsequently degraded by a DNase step. The addition of the 5' cap and the 3' poly(A) tail can be achieved during IVT or enzymatically after transcription.
