Effects of croton schiedeanus extract on nitric oxide production in the preparation of isolated aortic rings

En este trabajo se determinó la importancia de la ruta metabólica del óxido nítrico y la guanilato ciclasa en el efecto vasodilatador inducido por el extracto de Croton Schiedeanus Schlecht utilizando la preparación de anillos aislados de aorta de ratas Wistar. El extracto de C. Shiedeanus relaja anillos precontraídos con fenilefrina, agonista del receptor adrenérgico alfa, en función de la dosis, con una CE50 de 1,3x10-5 g/mL (9,1x10-6 – 2x10-5). Este efecto es superior al ejercido sobre anillos precontraídos con KCl (activador de canales de calcio dependientes de voltaje) y se reduce, pero no se suprime, en presencia de anillos sin endotelio. Azul de metileno (10-4 M), inhibidor de la guanilato ciclasa y L-NAME (10-4 M), inhibidor de la óxido nítrico sintasa, revierten completamente la relajación inducida por el extracto, en tanto que ésta no se afecta en presencia de propranolol (10-6 M), antagonista del receptor beta, de atropina (3x10-5 M), antagonista muscarínico e indometacina (10-6 M), inhibidor de ciclooxigenasa. Esto señala que la relajación inducida por C. Schiedeanus no está vinculada a la activación del receptor beta, ni a la activación del receptor muscarínico ni tampoco a la inducción de prostaglandinas. Adicionalmente, C. Schiedeanus no afecta las contracciones inducidas por CaCl2 lo que confirma que el antagonismo de calcio no es el mecanismo de relajación vascular de esta especie. sobre la ruta guanilato ciclasa/GMPc que sobre la actividad misma del óxido nítrico. Dado el interés de la actividad relajante de esta especie y la importancia de la ruta metabólica del NO/GMPc, no solo en el campo de la hipertensión sino en el de la aterogénesis y la agregación plaquetaria, los resultados de este estudio dan cuenta de que C. Schiedeanus podría constituirse en una fuente efectiva para la obtención de un fitoterapéutico con aplicaciones cardiovasculares. / Abstract. In this study we determined the importance of the metabolic pathway of nitric oxide and guanylate cyclase in the vasodilator effect induced by the extract of Croton schiedeanus Schlecht using the preparation of isolated aortic rings of Wistar rats. C, Shiedeanus relax rings precontracted with phenylephrine, alpha-adrenergic receptor agonist, depending on the dose, with an EC50 of 1.3 x10-5 g / mL (9.1 x10-6 - 2x10-5). This effect is superior to that exercised over rings precontracted with KCl (calcium channel activator of voltage-dependent) and is reduced but not abolished, in the presence of rings without endothelium. Methylene blue (10-4 M), an inhibitor of guanylate cyclase and L-NAME (10-4 M), an inhibitor of nitric oxide synthase, completely reversed the relaxation induced by the extract, while it is not affected in presence of propranolol (10-6 M), beta receptor antagonist, atropine (3x10-5 M), muscarinic antagonist and indomethacin (10-6 M), an inhibitor of cyclooxygenase. This indicates that the relaxation induced by C. Schiedeanus is not linked to activation of the beta receptor, or muscarinic receptor activation nor the induction of prostaglandins. Additionally, C. Schiedeanus not affect contractions induced by CaCl2 confirming that the antagonism of calcium is not the mechanism of vascular relaxation of this species. On the other hand, when testing serial dilutions of L-NAME and methylene blue to the order of 10-14M can be seen that the latter agent is more effective in reversing the relaxation induced by C. Schiedeanus, since in such conditions persist significant inhibitory effects (68% of maximal relaxation), whereas with L-NAME-induced relaxation of the extract is recovered well (91%). These findings suggest that C. Schiedeanus exert more selective effects on the route guanylate cyclase / cGMP that on the very activity of nitric oxide. Given the interest of the relaxant activity of this species and the importance of the metabolic pathway of NO / cGMP, not only in the field of hypertension but of atherogenesis and platelet aggregation, the results of this study relates to C. Schiedeanus could become an effective source for obtaining a phytotherapy with cardiovascular applications.Maestrí
