Cette thèse traite du développement et de l'utilisation d'outils microfluidiques pour l'encapsulation et l'analyse de cellules uniques dans le cadre d'applications en biologie et en cancérologie. Nous y relatons la mise en place et le développement d'une plateforme de microfluidique en gouttes composée de plusieurs modules permettant : l'encapsulation et le tri de cellule unique dans des gouttes aqueuses, l'injection et le mélange des cellules avec des réactifs et enfin l'analyse de leur contenu par un système de PCR quantitative en gouttes.
