Apoptosis is a genetically programmed, biochemically executed form of cell suicide essential for development and homeostasis in all multicellular organisma. The caspase family of cysteine proteases orchestrate the apoptosis program and are regulated by memebers of the Bcl-2 family. Extensive studies in mammals have revealed that Bcl-2 proteins control caspase activiation by governing a key mitochondrial checkpoint, invovling the permeabilization of the outer mitochondrial membrane and the release of proapoptotic factors from the intermembrane spce.
