At excitatory connections onto excitatory cells, NMDA receptors play a critical role in long-term plasticity. Upon glutamate binding, NMDA receptors allow the entry of calcium into postsynaptic compartments, which initiates a cascade of reactions leading to the strengthening of the connection.
