Human geranylgeranyl pyrophosphate synthase (hGGPPS) is an enzyme in the mevalonate pathway that catalyzes the formation of geranylgeranyl pyrophosphate (GGPP) via the condensation of farnesyl pyrophosphate (FPP) with isopentenyl pyrophosphate (IPP). GGPP is a key substrate for prenylation of a number of GTP binding regulatory proteins (G-proteins), including the Rho and Rac subfamilies that provide cell signalling. Biochemical studies have indicated a relationship between high levels of GGPP in the brain tissue and Alzheimer’s disease (AD) garnering an interest in targeting hGGPPS for the treatment of AD.
