Four groups of antibiotics (aminoglycosides, polymyxins, tetracyclines, and lincomycins) have neuromuscular blocking activity. The mechanism of neuromuscular blocking action of the most potent representative agents from each group was studied on isolated nerve and nerve-muscle preparations, and the predominant action was determined to be different for each of the groups. The predominant effect of neomycin, an aminoglycoside, on neuromuscular transmission is to reduce ACh release, and this effect results from blockade of calcium influx into the nerve terminals.
