The orientation of human erythrocyte membrane proteins was examined using the following techniques: i. lactoperoxidase-catalyzed iodination of the external surface with l25I and the cytoplasmic surface with 131I followed by proteolytic digestion of either the external surface (resealed ghosts) or cytoplasmic surface (inside-out vesicles), ii. phosphorylation of inside-out vesicles with alpha-32P-ATP in conjunction with alkaline phosphatase treatment; and iii. phospholipase C treatment of intact cells followed by enzymic iodination. Protein analysis by SDS polyacrylamide gel electrophoresis revealed several peaks of radioactivity whose orientation was as follows: i. a ~100,000 molecular weight component spanning the membrane asymmetrically; ...
