Spleen exonuclease and bacteriophage T4 induced polynucleotide kinase have been characterized with regard to their use in DNA sequence analysis and a model describing the action of polynucleotide kinase proposed. Interfering oligodeoxyribonucleotide-like contaminants in the polynucleotide kinase preparations were characterized and removed. A general method for the determination of the sequence of oligodeoxyribonucleotides of any base composition was developed. This involved partial spleen exonuclease degradation, 5' terminal labelling with polynucleotide kinase and [gamma-32P] ATP, and chain length fractionation of the labelled digest products.
